摘要
目的建立同时测定小檗碱甲氧苄啶胶囊中盐酸小檗碱和甲氧苄啶含量的HPLC法。方法采用Agilent ZORBAX SBC18色谱柱(250mm×4.6mm,5μm);流动相为乙腈-0.05mol·L^(-1)磷酸二氢钾(30∶70);进样量为20μL;柱温为30℃;流速为1.0mL·min-1;检测波长为270nm。结果盐酸小檗碱的线性范围为10.026 9~50.134 3μg·mL^(-1)(r=0.999 9),平均回收率为99.9%(RSD=0.3%,n=6);甲氧苄啶的线性范围为5.174 6~25.873 2μg·mL^(-1)(r=0.999 5),平均回收率为99.5%(RSD=0.8%,n=6)。结论该方法简便、快速、准确、重复性好,适用于小檗碱甲氧苄啶胶囊的质量控制。
Objective To establish an HPLC method for the determination of berberine hydrochloride and trimethoprim in Berberine Hydrochloride and Trimethoprim Capsules simultaneously. Methods Agilent ZORBAX SB-C 18 (250 mm×4.6 mm,5 μm) was used with acetonitrile and 0.05 mol·L -1 KH 2 PO 4 (30∶70) as the mobile phase and the flow rate was 1.0 mL·min -1 .The injection volume was 20 μL;the column temperature was set at 30 ℃,and the detection wavelength was set at 270 nm. Results The calibration curves for berberine hydrochloride and trimethoprim were linear over the range of 10.026 9-50.134 3 ( r =0.999 9) and 5.174 6 -25.873 2 μg·mL -1 ( r =0.999 5).The average recovery rates were 99.9% (RSD=0.3%, n =6),99.5% (RSD=0.8%, n =6),respectively. Conclusion This method is simple,rapid,accurate and reproducible,which is suitable for the quality control of Berberine Hydrochloride and Trimethoprim Capsules.
