摘要
目的从长白山特殊生境土壤中分离筛选出具有抗Staphylococcus aureaus与Escherichia coli两种临床常见指示菌效果明显的菌株。方法通过对菌株的初步分离,纯培养,革兰染色,抑菌实验,摇瓶发酵,形态特征等进行综合实验分析,优选具有较好抑菌作用的菌株。结果通过筛选,最终从16株形态典型的菌株中成功分离出2株具有明显抑菌效果的菌株,命名为D-x和D-f;菌株D-x与D-f发酵液上清及浸膏对两种靶标菌均具有明显的抑菌活性; D-x上清液对S.aureaus及E.coli的平均抑菌圈直径分别为13.8 mm和9.8 mm,浸膏对S.aureaus及E.coli的平均抑菌圈直径分别为10.5mm,7.2mm; D-f上清液对S.aureaus及E.coli平均抑菌圈直径分别达到12.5mm,10.8mm,浸膏对S.aureaus及E.coli平均抑菌圈直径分别达到9.6 mm,9.5 mm。结论通过筛选,菌株D-x与菌株D-f均具有较好抑菌活性,是两株值得开发的抗菌菌株。
Objective The aim was to isolate and screen the strains that had the obvious effect of anti Staphylococcus aureaus and Escherichia coli two clinical common target bacteria from the soil of Changbai Mountain special habitat.Methods A comprehensive analysis was conducted on the isolation,culture,and culture of the strain,and the inhibition test,the shape characteristic of the shaking flask and the morphol successfully isolated two strains has bacteriostasis effect strain,named for D-x,D-f and through further ogy of the strain.Results Through screening,ultimately from 16 strains of the typical morphology of strain shake flask fermentation,the fermentation supernatant and concentrated extract bacteriostasis experiment system.The experimental results showed that:strains D-x and D-f fermentation supernatant and extract has significant antibacterial activity on two kinds of target pathogens.D-x supernatant on S.aureaus and E.coli average inhibition zone diameter reached 13.8 mm and 9.8 mm,extract of S.aureaus and E.coli average inhibition zone diameter respectively reached 10.5 mm and 7.2 mm.D-f supernatant on S.aureaus and E.coli average inhibition zone diameter reached 12.5 mm,10.8 mm,extract of S.aureaus and E.coli average inhibition zone diameter reached 9.6 mm and 9.5 mm.Conclusion The strain D-x and strain D-f had good antibacterial activity,and it was a worthy of the development.
