摘要
目的:研究贵州苗药吉祥草(Reineckia camea)中的化学成分,以期能为吉祥草药理活性的阐明以及吉祥草药材的进一步开发利用提供科学依据。方法:取吉祥草全草药材20 kg,用80%乙醇加热回流提取3次,提取时间分别为2,2,1 h,将3次提取液合并,滤过,减压浓缩成浸膏。浸膏加适量的蒸馏水溶解后,依次用石油醚、乙酸乙酯、水饱和的正丁醇多次萃取,各部分萃取液经减压浓缩,干燥,得石油醚部位浸膏28 g,乙酸乙酯部位浸膏305 g,正丁醇部位浸膏812 g。对乙酸乙酯和正丁醇部位浸膏,利用硅胶柱,大孔树脂,小孔树脂(MCI)柱,ODS中压柱,LH-20型羟丙基葡聚糖凝胶(Sephadex LH-20)及Pre-HPLC等分离手段进行分离纯化,并通过理化性质,HR-ESI-MS,核磁波谱及文献数据进行结构鉴定。结果:从吉祥草的乙酸乙酯和正丁醇部位分离得到13个化合物,分别为2α,3β-dihydroxy-5α-pregn-16-en-20-one(1),(25R)-26-hydroxycholesterol(2),薯蓣皂苷元(3),β-谷甾醇(4),β-胡萝卜苷(5),异鼠李素-3-O-β-D-芸香糖苷(6),杜鹃素(7),木犀草素(8),1,2,8-trihdroxy-5,6-dimethoxyxanthone(9),獐牙菜苦苷(10),二(2-乙基己基)邻苯二甲酸酯(11),交链孢酚单甲醚(12),N-反式阿魏酸酪酰胺(13)。结论:化合物1,2,6~10,12,13均为首次从吉祥草中分离得到。
Objective: To study the chemical constituents from Guizhou Miao medicine Reineckia carnea,and provide a scientific basis for explaining the pharmacological activities of R. carnea and its further development and utilization. Method: The 20 kg air-dried whole plant of R. carnea was extracted with 80% Et OH for three times by heating reflux,2,2 and 1 h respectively. These 3 extracts were combined,filtered,decompressed and concentrated into extract,which was then dissolved in water. Then the concentrated residue was successively partitioned with petroleum ether,Et OAc and n-Bu OH to obtain 28 g petroleum ether fraction,305 g Et OAc fraction and 812 g n-Bu OH fraction. The chemical constituents of Et OAc and n-Bu OH fractions were isolated and purified by repeated silica gel column chromatography, macroporous resin, MCI, ODS, Sephadex LH-20 gel column chromatography and Pre-HPLC, and their structures were identified on the basis of physical and chemical properties,HR-ESI-MS,NMR spectral analysis and literature data. Result: Thirteen compounds were isolated and identified as 2α,3β-dihydroxy-5α-pregn-16-en-20-one(1),(25 R)-26-hydroxycholesterol(2),diosgenin(3),β-sitosterol(4),β-daucosterol(5),isorhamnetin-3-O-rutinoside(6),farrerol(7),luteolin(8),1,2,8-trihdroxy-5,6-dimethoxyxanthone(9),swertiamarin(10),di(2-ethylhexyl) phthalate(11),alternariol monomethyl ether(12),and N-trans-feruloyltyramine(13). Conclusion: Compounds 1,2,6-10,12,and 13 were isolated from this plant for the first time.
