Effects of Wnt-β-catenin signaling pathway on survival,proliferation,axon growth and synapse formation of mouse Schwann cells in vitro

摘要 目的检测Wnt/β-catenin信号通路对体外小鼠施万细胞存活、增殖及轴突生长和突触形成过程的影响。方法将施万细胞进行不同的分组,通过台盼蓝实验、CCK-8实验、共培养等实验检测Wnt通路对细胞增殖等功能的影响。结果加入Wnt抑制剂的分组,无论是施万细胞的存活率、增殖率、还是轴突生长程度和对照组相比显著降低。结论 Wnt/β-catenin信号通路影响体外小鼠施万细胞存活、增殖及轴突生长和突触形成过程。 Objective To detect the influence of the Wnt/beta-catenin signaling pathway on cell survival,proliferation,axon growth and synapse formation on the schwann cells of mouse in vitro. Methods These schwann cells would be divided into different groups,and the impact of Wnt signaling pathway on cell proliferation and other function would be detected via trypan blue assay,CCK-8,co-culture and some other assay methods. Results Compared with the control group,the group treated with Wnt inhibitor,not only the rate of cell survival and cell proliferation,but also the growth of axon were all decreased in schwann cells. Conclusion Wnt/beta-catenin signaling pathway affects the process of cell survival,proliferation,axon growth and synapse formation on mouse schwann cells in vitro.
