MEF2A RNA干扰对apoE^(-/-)小鼠动脉粥样硬化斑块及血清Lp-PLA2、MCP-1表达的影响 被引量:4

摘要 目的:观察肌细胞增强因子2A(MEF2A)表达下降对载脂蛋白E基因敲除(apoE^(-/-))小鼠动脉粥样硬化(AS)斑块形成及脂蛋白相关磷脂酶A2(Lp-PLA2)、单核细胞趋化蛋白-1(MCP-1)表达的影响。方法:高脂喂养apoE^(-/-)小鼠以构建AS模型。45只模型小鼠被分为MEF2A RNAi组、阴性对照组(NC组)和对照组,每组15只。MEF2A RNAi组动脉局部斑块滴注慢病毒悬液,NC组动脉局部斑块滴注NC慢病毒悬液,对照组滴加生理盐水,术后继续高脂喂养,5周后进行组织学检查。采用ELISA和real-time PCR法检测血清和斑块局部MEF2A及Lp-PLA2、MCP-1表达的变化。结果:MEF2A RNAi组与对照组、NC组相比,血清MEF2A水平降低,Lp-PLA2、MCP-1水平升高(P<0.05);AS斑块增厚,面积增大,斑块局部MEF2A mRNA表达水平降低,Lp-PLA2、MCP-1 mRNA表达水平增高(P<0.05)。结论:在体内apo E缺失时,抑制MEF2A活性可增强Lp-PLA2、MCP-1表达,促进AS及斑块形成。 Aim: To observe the effects of myocyte enhancer factor 2 A( MEF2A) RNA interference( RNAi) on atherosclerosis( AS) plaque formation and serum lipoprotein associated phospholipase A2( Lp-PLA2) and monocyte chemotactic protein 1( MCP-1). Methods: The apolipoprotein E-deficient( apoE-/-) mice were fed with a high-fat diet to induce AS model. A total of 45 mice were randomly allocated into three groups,each group of 15 mice: control group,lentivirus negative control group( NC group) and MEF2A RNAi group. Lentivirus suspension was dripped into the local plaque in MEF2A RNAi group,NC virus suspension was dripped into the local plaque in NC group,and physiological saline was dripped in the control group. High-fat feeding was continued,and 5 weeks later histological change was observed. ELISA and realtime PCR method were used to detect the MEF2A,Lp-PLA2 and MCP-1 expression in serum and local plaque. Results:Compared with the control group and NC group,the serum level of MEF2A decreased,and Lp-PLA2,MCP-1 increased( P 0. 05). The atherosclerotic plaque in the MEF2A RNAi group was significantly enlarged and thickened,the expression of local MEF2A mRNA significantly decreased,and the expressions of Lp-PLA2 and MCP-1 mRNA significantly increased( P 0. 05). Conclusion: In the absence of apo E,MEF2A RNAi could inhibit the activity of MEF2A,enhance the expression of Lp-PLA2 and MCP-1,and promote AS and plaque formation.
