摘要
目的:考察人参皂苷Rg_1对糖基化末端产物(AGEs)诱导炎症的影响及其潜在的作用机制。方法:MTT法评价人参皂苷Rg_1的大鼠肾小球系膜细胞(HBZY-1)毒性;2.5μg·ml^(-1)AGEs诱导HBZY-1细胞炎症,巨噬细胞浸润法评价人参皂苷Rg_1的抗炎作用;Western-Blot(WB)法探查人参皂苷Rg_1对TGF-β/Smad信号通路、还氧化酶-2(COX-2)与肿瘤坏死因子-α(TNF-α)表达量的影响。结果:人参皂苷Rg_1给药剂量≤10^(-6)mol·L^(-1)时,对HBZY-1细胞的生长无明显抑制作用。人参皂苷Rg_1抗炎作用的半数有效浓度为10^(-8)mol·L^(-1),最佳抗炎作用时间为24 h。巨噬细胞浸润实验中,10^(-8)~10^(-6)mol·L^(-1)人参皂苷Rg_1给药24 h对RAW 264.7细胞浸润诱发的炎症均有一定的抑制作用,10^(-7)和10^(-6)mol·L^(-1)人参皂苷Rg_1组与模型组比较吸光度显著降低(P<0.01)。WB实验中,10^(-6)mol·L^(-1)人参皂苷Rg_1可显著升高p-Smad2/3蛋白表达量,与模型组比较差异有统计学意义(P<0.05)。结论:人参皂苷Rg_1在一定给药浓度下具有抗炎作用,且该作用可能依赖于TGF-β/Smad信号通路活性的调节。
Objective: To investigate the effect of ginsenoside Rg1 on the inflammation induced by advanced glycation end products (AGEs) and the underling mechanism. Methods: HBZY-1 cytotoxicity of ginsenoside Rg1 was evaluated by MTT. The inflammation was induced by 2.5 μg·ml^-1 AGEs, and then the effects of ginsenoside Rg1 on TGF-β/Smad signaling pathway, cyelooxygenase-2 ( COX-2 ) and tumor necrosis factor-α (TNF-α) were evaluated by Western-blot (WB). Results: When the dosage of ginsenoside Rg1 was tow or equal to 10^-6 mol·L^-1 , there was no obvious inhibitory effect on HBZY-1 cell. The EC50 of anti-inflammation was 10^-8 mol·L^-1 and the best treatment period was 24 h. In the maerophage infiltration test, 10^-8-10^-6 mol·L^-1 ginsenoside Rg1 could inhibit the inflammation induced by RAW 264.7 cell in 24 h. The absorbance of 10^-7 and 10^-6 mol·L^-1 ginsenoside Rg1 group was sig- nificantly lower than that of the model group ( P 〈 0.01 ). In the WB experiment, 10-6 mol·L^-1 ginsenoside Rg1 significantly in- creased the expression of p-Smad2/3 protein, and when compared with model group, the difference was statistically significant (P 〈 0.05 ). Conclusion: Ginsenoside Rg1 plays a role in anti-inflammation via regulating TGF-β/Smad signal pathway activity.
