跑台运动对骨质疏松大鼠骨组织Wnt/β-catenin信号通路的影响 被引量:10

Treadmill exercise can significantly improve bone metabolism in osteoporosis

摘要 目的研究跑台运动对骨质疏松大鼠骨代谢的影响以及Wnt/β-catenin信号通路在运动影响骨代谢中的调节作用。方法选取2月龄SD雌性大鼠32只,按随机数字表法分为假手术安静组(S组)、去卵巢安静组(O组)、假手术运动组(ST组)和去卵巢运动组(OT组),每组大鼠8只。采用去卵巢手术制造大鼠绝经后骨质疏松模型,O组和OT组均摘除卵巢,S组和ST组大鼠的手术只进行到找到卵巢,但并不摘除卵巢。造模成功后,S组和O组大鼠在饲养笼中自由活动,ST组和OT组大鼠除在饲养笼中自由活动外.每Et还进行跑台训练。于ST组和0T组最后1次运动结束24h后(人组10周后)处死4组大鼠,分别采用双能X线骨密度仪测定大鼠股骨骨密度(BMD),全自动生化分析仪检测血清钙(Ca)、磷(P)、碱性磷酸酶(ALP)含量,放射免疫法检测血清骨钙素(BGP)活性,三点弯曲试验检测股骨生物力学指标,WeSternblot测定骨组织Wnt/β-catenin信号通路相关蛋白的表达。结果人组10周后,O组大鼠BMD显著低于S组大鼠,ST组大鼠BMD显著高于S组大鼠,OT组大鼠BMD显著高于O组大鼠BMD,组间差异均有统计学意义(P〈O.05);ST组大鼠Wnt/13.catenin信号通路蛋白P-GSK.313的相对表达量为(0.51±0.06),显著低于S组的(0.80,0.05),组间差异有统计学意义(P〈0.05),而ST组大鼠β-catenin和eyelinD1的相对表达量均显著高于S组,组间差异均有统计学意义(P〈0.05);0T组大鼠Wnt/13.catenin信号通路蛋白P.GSK-3β的相对表达量显著低于O组,而β-catenin和cyclinD1的相对表达量则显著高于O组,组间差异均有统计学意义(P〈0.05)。结论跑台运动可显著改善骨质疏松大鼠的骨代谢,其机制可能与Wnt/β-eatenin信号通路的激活相关。 Objective To study the effect of treadmill exercise on the bone metabolism of osteoporotic rats and the regulatory role of Wnt/β-catenin signaling in it. Methods Thirty-two female Sprague-Dawley rats aged 2 months were randomly divided into a sham-operated quiet group (S group ), an ovariectomized quiet group (O group), a sham-operated exercise group (ST group) and an ovarieetomized exercise group (OT group) with 8 rats in each group. The ovariectomies established a model of post-menopausal osteoporosis. The ovaries of groups O and OT were removed, while those of the S and ST groups were exposed, but not removed. All of the rats were free to move in their cages, but those in the ST and OT groups also underwent treadmill training every day for 10 weeks. All of the rats were sacrificed 24 hours after the last exercise session. Bone mineral density (BMD) of the femur was measured using dual energy X-ray absorptiometry, while serum calcium, phosphorus and alkaline phosphatase were detected with a biochemical analyzer. Serum osteocalcin was quantified by radioimmunoassay. A three point bending test quantified the biomeehanical properties of the femors. The expression of Wnt/β-eatenin was measured through western blotting. Results After the intervention the average BMD of group O was significantly lower than that of group S, that of. group ST was significantly higher than group S and that of group OT significantly higher than group O (P ≤0.05). The relative expression of Wnt/β-catenin protein P-GSK-3β in group ST was significantly lower than in group S (P ≤ 0.05). The relative expressions of β-eatenin and cyelin D1 in group ST were significantly higher than in group S (P ≤0.05). The relative expression of protein P-GSK-3βin group OT was significantly lower than in group O, while that of β-catenin and cyclin D1 was significantly higher (P ≤0.05). Conclusion Treadmill exercise can significantly improve bone metabolism in osteoporotic rats. The mechanism may be related to activating the Wnt/β-catenin signaling pathway.
