摘要
目的测定妊娠期糖尿病(gestational diabetes mellitus,GDM)及正常妊娠妇女胎盘组织中丝氨酸蛋白酶抑制剂(vaspin)及白细胞介素-6(interleukin-6,IL-6)的表达,观察IL-6对Be Wo细胞vaspin表达的影响,探讨vaspin、IL-6在GDM中的作用及相互关系。方法选择2012年10月至2013年10月于河北省人民医院行择期剖宫产术57例孕妇,根据孕妇有无GDM分为GDM组(30例)和正常妊娠(normal glucose tolerance,NGT)组(27例)。取两组孕妇胎盘组织,实时荧光定量反转录PCR测定胎盘组织vaspin、IL-6mRNA表达水平,免疫蛋白印迹法检测胎盘组织vaspin及IL-6蛋白的表达水平;用不同浓度的IL-6分别与Be Wo细胞共培养,留取细胞,测定Be Wo细胞vaspin mRNA和蛋白表达水平。结果 GDM组胎盘组织vaspin mRNA及蛋白表达水平与NGT组比较[(0. 60±0. 32) vs (0. 68±0. 32);(0. 30±0. 08) vs (0. 39±0. 26)],差异无统计学意义(P> 0. 05); IL-6 mRNA及蛋白表达水平高于NGT组[(1. 35±0. 67) vs (0. 82±0. 47);(1. 23±0. 25) vs (0. 54±0. 20)](P <0. 05); GDM组胎盘组织vaspin mRNA和蛋白表达与新生儿体质量呈负相关(r=-0. 59、-0. 88,P=0. 01、<0. 01)。不同浓度的IL-6均可抑制Be Wo细胞vaspin mRNA及蛋白表达。结论 vaspin与GDM的发生有关,并参与胎儿生长发育; IL-6可能参与了vaspin的调控。
Objective To investigate the mRNA and protein expression of vaspin and interleukin-6( IL-6) in the placenta tissues in pregnant women with or without gestational diabetes mellitus( GDM),and to observe the changes of the vaspin expression on Be Wo cells after cultured with different concentrations of IL-6,analyzing their correlationship and roles in GDM. Methods A total of57 patients who underwent elective cesarean section in People's Hospital of Hebei Province from October 2012 to October 2013 were selected and divided into GDM group and normal glucose tolerance( NGT) group. The placental tissues of the two groups were taken.The expression levels of vaspin and IL-6 mRNA in placenta were detected by real-time fluorescent quantitative reverse transcription-PCR. The expression levels of vaspin and IL-6 protein in placenta were detected by immunoblotting. Different concentrations of IL-6 were used to co-culture with Be Wo cells,and determine the expression level of vaspin mRNA and protein in Be Wo cells. Results The expression of vaspin mRNA and protein in placenta of GDM group was compared with those of NGT group [( 0. 60 ± 0. 32) vs( 0. 68 ± 0. 32);( 0. 30 ± 0. 08) vs( 0. 39 ± 0. 26) ],the differences were not statistically significant( P〈0. 05). The expression levels of IL-6 mRNA and protein were higher than those in the NGT group [( 1. 35 ± 0. 67) vs( 0. 82 ± 0. 47);( 1. 23 ± 0. 25) vs( 0. 54 ± 0. 20) ]( P〈0. 05). The expression of vaspin mRNA and protein in placenta of GDM group was negatively correlated with the body weight of newborns( r =-0. 59,-0. 88; P = 0. 01, 0. 01). Different concentrations of IL-6 can inhibit the expression of vaspin mRNA and protein in Be Wo cells. Conclusion Vaspin is involved in the development of GDM and participates in fetal growth and development; IL-6 may be involved in the regulation of vaspin.
