逍遥合剂对产后抑郁症模型大鼠行为学、神经递质及ERK1/2通路影响的研究 被引量:10

摘要 目的:探讨逍遥合剂对产后抑郁症模型大鼠行为学、神经递质及ERK1/2通路的影响。方法:SPF级雌性SD大鼠共72只,随机分正常组、模型组(生理盐水1ml/100g)、逍遥合剂高、中、低剂量组即7、3.6、1.8g(kg·d)、阳性药组用盐酸氟西汀组10mg/(kg·d),每组有12只。各模型组肌注苯甲酸雌二醇及黄体酮以建立产后抑郁症模型大鼠,共造模23d。造模成功后予各治疗组灌胃药物,正常组和模型组灌胃生理盐水,观察各组大鼠体重、行为学、神经递质及ERK1/2蛋白表达情况。结果:3周后,各治疗组大鼠体重均高于模型组体重,且正常组、逍遥合剂高、中剂量组显著高于模型组(P<0.05),逍遥合剂的高剂量组体重显著高于盐酸氟西汀组(P<0.05)。治疗前各治疗组大鼠行为学指标评分均无显著差异(P>0.05),治疗后逍遥合剂高、中剂量组及盐酸氟西汀组的水平移动总路程较模型组有统计学意义(P<0.05),逍遥合剂高、中、低剂量组及盐酸氟西汀组的中央区域路程较模型组有统计学意义(P<0.05)。逍遥合剂高、中剂量组5-HT、DA、NE水平较模型组显著上升(P<0.05)。逍遥合剂高、中剂量组p-CREB、p-ERK1/2蛋白水平较模型组显著上升(P<0.05),逍遥合剂低剂量组及盐酸氟西汀组较逍遥合剂高剂量组显著降低(P<0.05)。结论:逍遥合剂能有效改善产后抑郁模型大鼠行为学各项指标,可能是通过上调下丘脑神经递质及ERK1/2信号通路上p-MEK1、MEK1蛋白水平来缓解抑郁症状。 Objective: To investigate the effects of Xiaoyao mixture on behavior, neurotransmitter and ERK1/2 pathway in postnatal depression rats. Medthods: A total of 72 SPF female SD rats were randomly divided into normal group, model group(Normal saline 1ml/100g), Xiaoyao mixture high, middle and low dose group 7, 3.6, 1.8g/(kg·d) and positive drug group fluoxetine hydrochloride group 10 mg/(kg·d) with 12 rats in each group. The rats in each group were injected with estradiol benzoate and progesterone to establish postnatal depression model rats for 23 days. The normal group and the model group were given the normal saline, and the body weight, behavior, neurotransmitters and ERK1/2 protein expression were observed in the normal group and the model group. Results: After 3 weeks, the weight of the rats in the treatment group was higher than that of the model group, and the normal group, the Xiaoyao mixture high and middle dose group were significantly higher than the model group ( P 〈0.05). The high dose group of Xiaoyao mixture was significantly higher than that of the fluoxetine hydrochloride group ( P 〈0.05). Before treatment, there was no significant difference ( P 〉 0.05). After treatment, the total distance movement of Xiaoyao mixture high, middle dose group and fluoxetine hydrochloride group was significantly different than that of the model group ( P 〈0.05). The central regional route of Xiaoyao mixture high, middle and low dosage groups and fluoxetine hydrochloride group was more than that of the model group. There were significant differences in the model group ( P 〈0.05). The levels of 5-HT, DA and NE in the high and middle dose groups of Xiaoyao mixture were significantly higher than those in the model group ( P 〈0.05). The level of p-CREB and p-ERK1/2 protein in the high and middle dose groups of Xiaoyao mixture was significantly higher than that in the model group ( P 〈0.05). The low dose group and the high dose group of the fluoxetine hydrochloride group were significantly lower than those in the high dose group ( P 〈0.05). Conclusion: Xiaoyao mixture can effectively improve the behavior of postpartum depression model rats, possibly by raising the level of p-MEK1 and MEK1 protein in the hypothalamus neurotransmitter and ERK1/2 signaling pathway to alleviate the depressive symptoms.
