双参通脉颗粒对大鼠心肌缺血再灌注损伤的保护作用 被引量:5

摘要 目的:研究双参通脉颗粒对大鼠心肌缺血的可能保护机制。方法:SD大鼠按随机数字表法分为假手术组(Sham)、模型组(MIRI)、西药组(DH)、中药低、中、高剂量组(GSG-L、GSG-M、GSG-H),灌胃4周结扎左冠状动脉前降支造模,测定心功能、病理变化、谷胱甘肽过氧化酶(GSH-PX)、过氧化氢酶(CAT)、内皮素(ET)、髓过氧化物酶(MPO)含量变化,原位末端转移酶标记技术(TUNEL)染色评估心肌细胞凋亡指数,Western检测心肌核转录因子-κB(NF-κB)、Iκκβ蛋白表达。结果:DH组、GSG各剂量组与MIRI组比较,心功能改善明显,DH组、GSG各组血清ET、MPO水平、心肌NF-κB蛋白表达明显低于MIRI组,GSH-PX、CAT水平、心肌Iκκβ蛋白表达明显高于MIRI组。结论:GSG可抑制大鼠心肌缺血氧化损伤,其机制可能与减少ET、MPO含量,抑制NF-κB蛋白表达,增加GSH-PX、CAT含量,提高Iκκβ蛋白表达有关。 Objectine: To explore the protective effects of GSG on myocardial ischemia oxidative damage in rats. Methods:SD rats were divided into Sham operation group(Sham), myocardial ischemia reperfusion injury group(MIRI), diltiazem hydrochloride group(DH) and ginseng and salvia granules for promoting blood flow group of low dose, middle dose(GSG-M) and high dose(GSG-H). The rats were fed with saline and DH, GSG for four weeks. Then ligatured the left anterior descending coronary artery, the contents of GSH-PX、CAT、ET、MPO in plasma were measured after administration. Td-mediated dUTP nick end labeling(TUNEL) staining to assess the apoptosis index of myocardial cells. The NF-κB and Iκκβ protein levels were measured by the techniques of Western. Results: Compared to MIRI group, the cardiac diastolic function was significantly improved in GSG each dose and DH treatment group, the plasma levels of ET、MPO were lower, the plasma levels of GSH-PX、CAT were higher. The results of Western blot showed that the protein expressions of NF-κB were lower in GSG each dose group than those in MIRI group,the protein expressions of Iκκβ were higher in GSG each dose group than those in MIRI group, the high dosage group has better curative effect. Conclusion: GSG can alleviate myocardial ischemia oxidative damage in rats by decreasing NF-κB expression protein levels and improving Iκκβ expression protein levels.
