GluN3A型NMDAR在三氟淫羊藿素抗脑缺血再灌注损伤中的作用研究

摘要 目的:研究三氟淫羊藿素(Icaritfluor,ICTF)对脑缺血再灌注损伤的保护作用,初步探讨ICTF对GluN3A mRNA水平和蛋白表达的影响。方法:采用大鼠大脑中动脉栓塞(MACO)动物模型,TTC染色测定脑梗死范围,q-PCR方法检测GluN3A mRNA的表达,Western blot方法检测GluN3A蛋白表达水平。结果:MCAO再灌注模型组大鼠出现明显的脑梗死,缺血半损伤区脑组织GluN3A mRNA及蛋白表达均降低,给予ICTF后,脑梗死体积缩小,GluN3A mRNA及蛋白表达均升高。结论:ICTF对脑缺血再灌注损伤具有保护作用,其保护作用可能与升高NMDA受体的GluN3A亚基表达有关。 Objective: To study the protective effect of Trifluoro-icaritin (ICTF) on cerebral ischemia/reperfusion injury(I/RI), and investigate the mRNA and protein expression of GluN3A content N-methyl-D-aspartate receptor (NMDAR). Methods: The MCAO animal model in Sprague-Dawley rat was adopted, and the cerebral infarction range was determined by TTC staining. The expression of GluN3A mRNA and protein were detected by q-PCR and Western blot assay respectively. Results: The ischemia/reperfussion(I/R) model group showed an obvious cerebral infarction and decreases expression of GluN3A mRNA and protein in the ischemic semi-injury area. The volume of cerebral infarction was reduced and the expression of GluN3A mRNA and protein were increased after ICTF treatment. Conclusion: ICTF has an inhibition effect on cerebral I/R injury, which may be relevant with the increasing of GluN3Acontent NMDAR.
