摘要
以西洋梨(Pyrus communis L.)品种‘红考密斯’新生长的半木质化嫩枝茎段为外植体,促使腋芽萌发获得无菌试管苗;以试管苗为试材,研究基本培养基、外源植物生长物质对增殖和生根的影响;以试管苗的离体叶片为外植体,研究植物生长物质的种类和浓度对叶片不定梢再生的影响。结果表明,试管苗适宜的增殖培养基为MS+0. 5 mg·L^(-1)6-BA+0. 1 mg·L^(-1)TDZ+0. 3 mg·L^(-1)IBA;适宜的壮苗培养基为MS+0. 1mg·L^(-1)TDZ+0. 3 mg·L^(-1)IBA;适宜的生根培养基为1/2MS(仅MS大量元素减半)+0. 3 mg·L^(-1)NAA+20 g·L^(-1)蔗糖,生根率为82. 2%,平均单株根数为3. 3条;叶片不定梢再生的最佳培养基为:NN69+2 mg·L^(-1)6-BA+0. 3 mg·L^(-1)IBA,不定梢再生率为76. 8%,平均每叶不定梢数为1. 2个。
Young semi-lignified branches were selected as explants, and axillary buds were promoted to germinate and to form in vitro shoots. In vitro shoots were selected as materials, the effects of basic medium and external plant growth substance on proliferation and rooting were examined. In vitro leaves were selected as explants, and the effects of the kinds and concentrations of plant growth substances on shoot regeneration were investigated.The results showed that the proper proliferation medium was MS+0.5 mg·L -1 6-BA+ 0.1 mg· L -1 TDZ+0.3 mg·L -1 IBA and the proper medium for healthy shoots was MS+0.1 mg·L -1 TDZ+ 0.3 mg·L -1 IBA. The proper rooting medium was 1/2MS+0.3 mg·L -1 NAA+20 g·L -1 sucrose, the highest rooting rate was 82.2%, and the average root number per plantlet was 3.3. The optimal shoot regeneration medium was NN69+2 mg·L -1 6-BA+0.3 mg·L -1 IBA, the shoot regeneration frequency was 76.8%, and the average shoot number per leaf was 1.2.
