摘要
本研究以晚熟杂柑清见(Kiyomi tangor)为研究材料,测定了其果实发育成熟七个时间点的果肉及叶片中IAA的含量,并通过qRT-PCR分析了miR167及其靶基因ARF8的表达情况。结果,发现清见果肉IAA含量在果实膨大期和转色期出现一个双峰的动态变化,叶片中IAA含量在花后180 d上升到达最大值后下降,表明叶片IAA可能在果实转色期向果肉转移,参与果实着色的相关代谢;果肉中ARF8在花后90 d前的表达出现显著上调时miR167以及miR167a/miR167b/miR167c的表达出现显著下调,叶片中ARF8在花后90 d显著下调时miR167以及miR167a/miR167b/miR167c显著上调,表明此时果肉及叶片内miR167以及miR167a/miR167b/miR167c与靶基因ARF8发生了相互抑制作用,此后miR167及miR167a/miR167b/miR167c表达出现变化时ARF8表达量几乎都保持在一个很低的水平,表明这些miRNA可能参与了其他基因的调控。
In this study, Kiyomi tangor, one of the late maturing Citrus, was used as the research material to determine the content of IAA in the pulp and leaves at seven points of time during the fruit riperfing stage of Kiyomi tangor, In addition, qRT-PCR was used to analyze the expression 0fmiR167 and its target gene ARF8. The results showed that the IAA content in the pulp had a bimodal dynamic change during the period of fruit enlargement and turning-color and the content of IAA in leaves increased to the maximum at 180 d after flower booming and then decreased, indicating that IAA in leaves might transfer to the pulp in the fruit turning-color period and participate in metabolism related to fruit coloration. Within 90 d aiter flowering, the expression of A RF8 in the pulp was significantly up-regulated, while the expression of miR167 and miR167a/miR167b/miR167c was down regulated significantly in the meantime. Moreover, the expression of ARF8 in leaves at 90 d after flower booming was significantly down-regulated, but miR167 and miR167a/miR167b/miR167c were significantly up-regulated, which indicated that miR167 and miR167a/miR167b/miR167c in the pulp and leaves had mutal inhibition with the target gene ARF8 at this time. Hereafter, when the expression of miR167 and miR167a/ miR167b/miR167c changed, the expression of ARF8 remained at a very low level, which suggested that these miRNA might participate in the regulation of other genes.
