摘要
花青素还原酶(anthocyanidin reductase, ANR)是植物原花青素合成代谢途径中的关键酶之一,对植物类黄酮类物质的合成有重要作用。本研究以黔产艾纳香叶片为材料,根据其转录组测序结果,通过RT-PCR方法成功克隆得到艾纳香ANR基因的全长c DNA序列。随后,利用生物信息学在线工具对该ANR基因所编码蛋白的生理和化学参数、功能结构域、亲/疏水性、信号肽、二/三级结构等进行了相关预测和分析。分析结果表明,该基因cDNA全长为978 bp,编码氨基酸326个,所编码蛋白分子量为36.30 kD,等电点为5.83,为亲水性蛋白,不含信号肽序列。
Anthocyanidin reductase (ANR) is one of the key enzyme in the procyanidins biosynthetic pathway of plants, which is important for the synthesis of plant flavonoids. In this study, Blumea balsamifera leaves were used as research materials. According to the results of its transcriptome sequencing, the full-length cDNA sequence of Blurnea balsamifera ANR was cloned successfully by RT-PCR method. Then the physiological and chemical parameters, functional domains, hydrophobicity/hydrophilicity properties, signal peptide, secondary/tertiary structure and so on of the protein encoded by the A NR gene were predicted and analyzed by using bioinformatics online tools. The results showed that the full-length eDNA of ANR was 978 bp, and 326 amino acids were encoded. The encoded protein molecular weight was 36.30 kD and its isoelectric points were 5.83. ANR of Blurnea balsamifera was a hydrophilic protein, had no signal peptide sequence.
