姜黄素保护染镉大鼠肾脏功能研究

摘要 目的探讨姜黄素对染镉大鼠肾脏损伤的保护作用。方法选取8周龄健康Wistar大鼠48只,按照数字表法随机分为对照组、染镉组、姜黄素组,每组各16只。对照组腹腔注射生理盐水20 mg/kg,染镉组腹腔注射氯化镉溶液1.4 mg/kg,姜黄素组在染毒组基础上,皮下注射姜黄素50 mg/kg。各组均为每周5次,1次/d,连续注射5周。分别检测造模前后大鼠体质量、肾脏质量系数;24 h尿液、尿镉含量、β2微球蛋白;血镉含量、尿素氮(BUN)、血肌酐(Scr);肾皮质中超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)、肾皮质镉含量。结果与对照组比较,染镉组和姜黄素组大鼠体质量、肾脏质量系数明显降低;24 h尿量、β2微球蛋白、尿镉含量明显升高;血镉、BUN、Scr含量明显升高;肾皮质中SOD、GSH-Px活力明显降低,MDA、镉含量明显升高,差异有统计学意义(P<0.05)。与染镉组比较,姜黄素组大鼠体质量、肾脏质量系数明显升高,24 h尿量、β2微球蛋白、尿镉含量明显降低;血镉、BUN、Scr含量明显降低;肾皮质中SOD、GSH-Px活力明显升高,MDA、肾皮质镉含量明显降低,差异有统计学意义(P<0.05)。结论姜黄素明显保护了镉对大鼠肾功能的损害,抑制肾组织发生脂质过氧化反应。 Objective To explore the protective role of curcumin on kidney function of rats exposed to cadmium.Methods 48 healthy Wistar rats aged 8 weeks were selected, and were randomly divided into control group, exposure group and curcumin group according to the numerical table, with 16 rats in each group. The control group was given intraperitoneal injection of normal saline with dose of 20 mg/kg, the exposure group was given intraperitoneal injection of cadmium chloride solution with dose of 1.4 mg/kg, and the curcumin group was given subcutaneous injection of curcumin with dose of 50 mg/kg on the basis of the exposure group. Each group was given five consecutive weeks of injection once a week. Following indicators were measured: the weight and kidney mass coefficient of rats before and after modeling; urine, urine cadmium content and chloramine 2 microglobulin for 24 h; blood cadmium content, urea nitrogen(BUN), blood creatinine(Scr); the contents of superoxide dismutase(SOD), malondialdehyde(MDA), glutathione peroxidase(GSH-Px) and cadmium in renal cortex.Results Compared with the control group, the body weight and kidney mass coefficient were significantly reduced in the exposure group and the curcumin group. The contents of urine, chloramine and urine cadmium were significantly increased at 24 h. Blood cadmium content, BUN and Scr were significantly increased. In the renal cortex, SOD and GSH-Px were significantly decreased, while MDA and cadmium contents were significantly increased(P〈0.05). Compared with the exposure group, the body weight and renal mass coefficient of the curcumin group were significantly increased, and the urine volume, the content of chloramine 2 microglobulin and urine cadmium were significantly decreased at 24 h. The content of blood cadmium, BUN and Scr decreased significantly. The content of SOD and GSH-Px in renal cortex was significantly increased, while the content of MDA and cadmium in renal cortex was significantly decreased, with statistically significant differences(P0.05). Conclusions Significantly protected renal function in cadmium-exposed rats, inhibited lipid peroxidation in renal tissues.
