摘要
目的探讨血管生成素(Ang)-1促进骨髓基质干细胞(BMSCs)结合聚乳酸乙醇酸羟基磷灰石(PLGA/HA)支架治疗兔桡骨缺损的效果。方法选取健康3月龄新西兰大白兔48只,随机分为Ang-1基因转染BMSCs-PLGA/HA复合多孔支架组(A组)、无Ang-1基因转染组(B组)、单纯支架组(C组)和空白对照组(D组),每组12只。分别于支架植入兔体内1、4和8周取材,采用Micro CT检测缺损部位新生骨量,免疫组化法比较血管断面数目,记录比较碱性磷酸酶(ALP)、Ⅰ型胶原(Col I)、骨钙素(OC)和骨桥素(OPN)阳性表达。结果各组实验兔骨矿物质含量(BMC)均随时间延长而增加,且各时间点A组高于B组,C组次之,D组最小,差异均有统计学意义(均P <0. 05)。各组实验兔血管断面数目和ALP、Col I、OC及OPN阳性率均随时间延长而增多,且各时间点A组高于B组,C组次之,D组最少,差异均有统计学意义(均P <0. 05)。结论 Ang-1可促进BMSCs结合PLGA/HA支架修复兔桡骨缺损,促进新生骨形成和血管新生。
Objective To study the efficacy of Angiopoietin( Ang)-1 promoting bone mesenchymal stem cells( BMSCs) combined with poly lactic-co-glycolic acid/hydroxyapatite( PLGA/HA) stent for radial defect in rabbits. Methods A total of 48 three months old healthy New Zealand rabbits,were randomized to gene transfected BMSCs-PLGA/HA combined with porous scaffold( group A),non-Ang-1 transfection( group B),single scaffold group( group C) and a blank control group( group D) with 12 rabbits in each group. They were respectively transplanted in vivo into rabbits for 1 week, 4 weeks and 8 weeks. Amount of new bone formation were measured by Micro CT. The number of vascular sections were compared by immunohistochemistry and positive expression of alkaline phosphatase( ALP),type I collagen( Col I),osteocalcin( OC) and osteopontin( OPN) were also recorded and compared. Results Bone mineral content( BMC) increased as time goes on in each group with group A higher than group B followed by group C and then group D( all P〈0. 05). The number of vascular sections,positive expression of ALP,Col I,OC and OPN also increased as time goes on in each group with group A higher than group B followed by group C and then group D( all P〈0. 05). Conclusion Ang-1 can promote BMSCs combined with PLGA/HA scaffold for radial defect rehabilitation in rabbits,as well as promote new bone formation and angiogenesis.
