摘要
目的建立反相高效液相色谱法(RP-HPLC)测定忧遁草中夏佛塔苷含量,比较两个产地的忧遁草中夏佛塔苷的含量。方法采用Diamonsil C18(2)色谱柱(250 mm×4.6 mm,5μm),流动相A相为0.1%乙酸水溶液,B相为甲醇,等度洗脱:0~60 min 24%B,流量1 ml/min,柱温30℃,检测波长272 nm。结果夏佛塔苷在5.88~15.68μg范围内与峰面积呈良好的线性关系(R2=1);平均加样回收率为95.49%,RSD为1.61%。结论该方法简单,准确,重复性好,国产忧遁草中夏佛塔苷的含量明显地高于印度尼西亚忧遁草。
Objective To establish a RP-HPLC method for determination of schaftoside in Clinacanthus Nutans (Burm. F.) Lindau and compare the content of schaftoside in Clinacanthus Nutans (Burm. F.) Lindau from two origins. Methods A Diamonsil C18 (2) column (250 mm×4.6 mm, 5 μm) was adopted, with the temperature set at 30 ℃. The mobile phase consisted of 1 % acetic acid (A) and methanol (B). The isocratic elution was used according to the following profle: 0-60 min, 24 % B. The detection wavelength was 272 nm. Results The calibration curve of schaftoside had a good linearity within 5.88-15.68 μg (R2=1). The average recovery was 95.49 %, with RSD of 1.61 %. Conclusion The method is simple and accurate, with excellent repeatability. The content of chataroside in Clinacanthus Nutans (Burm. F.) Lindau from China is signifcantly higher than that in Indonesia.
