摘要
为探讨人参水提物对雄性果蝇的抗衰老作用机制,以Oregon K雄性黑腹果蝇为研究对象,将2日龄果蝇随机分为4组(对照组和不同剂量人参水提物组),每组200只,记录果蝇死亡数并统计平均寿命和平均最高寿命;测定氧化损伤条件下,人参水提物对果蝇存活时间的影响;测定超氧化物歧化酶(Superoxide dismutase,SOD)、过氧化氢酶(Catalase,CAT)活力和丙二醛(Malondialdehyde,MDA)含量;采用qRT-RCR法测定相关基因转录水平,阐明人参水提物抗衰老作用机制。试验结果表明:与对照组相比,人参水提物4 mg/m L和12 mg/m L剂量组果蝇平均寿命及平均最高寿命显著延长(P<0. 05或P<0. 01)。4 mg/m L和12 mg/m L人参水提物可在一定程度上延长氧化损伤状态下果蝇的存活时间(P<0. 05),提高SOD和CAT酶活力(P<0. 05或P<0. 01),降低MDA含量(P<0. 05),显著上调SOD和CAT mRNA表达水平(P<0. 000 1),下调MTH基因表达(P<0. 000 1)。这说明人参水提物可提高果蝇抗氧化能力,延缓果蝇衰老。
Taking Oregon K male Drosophila melanogaster as research object,2-day old Drosophila were randomly divided into 4 groups( control group and different-dose ginseng water extract groups),200 in each group,the number of deaths was recorded,and average life span and average maximum life span were calculated. The effects of water extracts of ginseng on survival time of Drosophila were determined under oxidative damage conditions. The activities of superoxide dismutase( SOD),catalase( CAT) and malondialdehyde( MDA) were measured. qRT-RCR method was used to determine the transcriptional levels of related genes,and the anti-aging mechanism of ginseng water extracts was elucidated. Results show that compared with the control group,the average life span and average maximum life span of Drosophila melanogaster were significantly prolonged in 4 and 12 mg/m L ginseng extracts( P〈0. 05 or P〈0. 01). 4 and 12 mg/m L ginseng extracts can ex-tend the survival time of Drosophila under oxidative damage to a certain extent( P〈0. 05). The activities of SOD and CAT were increased( P 0. 05 or P 0. 01) and the content of MDA was decreased( P〈0. 05). The expression levels of SOD and CAT mRNA were significantly up-regulated( P〈0. 000 1),and MTH( methuselah) gene expression was down-regulated( P〈0. 000 1). Research findings show the extracts of ginseng can improve the antioxidant capacity of Drosophila melanogaster and delay the aging of Drosophila melanogaster.
