摘要
采用高压脉冲电场(High voltage pulsed electric field,PEF)对五味子藤茎中木脂素的提取工艺进行了研究。以料液比、电场强度及脉冲数为自变量,以五味子醇甲(Schisandrol A)、五味子醇乙(Schisandrol B)、五味子甲素(Deoxyschisandrin)、五味子乙素(Schisandrin B)等4种木脂素的得率为考察指标,在单因素试验结果的基础上,通过3因素3水平响应曲面法(Response surface methology,RSM),对五味子藤茎中4种木脂素的PEF快速提取工艺方法进行优化研究。结果表明:以80%乙醇为提取溶剂,电场强度25 k V/cm,脉冲数为8,料液比1∶30 (g∶m L)是最佳提取工艺条件,其得率为2. 28%,该提取工艺提取速度快,经济、省时,适合工艺化生产。
High-voltage pulsed electric field( PEF) was used to study the extraction process of lignans in the stems of Schisandra chinensis. Taking ratio of material to liquid,electric field intensity and pulse number as dependent variables,yields of four kinds of lignans,including Schisandrol A,Schisandrol B,Deoxyschisandrin,Schisandrin B as indicators,rapid extraction process of the four lignans from lignans of Schisandra chinensis was optimized by 3-factor 3-level response surface methodology( RSM). The results showed that the yield is 2.28% with the optimum extraction conditions of 80% ethanol extraction solution,25 kV/cm electric field intensity,8 pulse number, and1 ∶ 30 g/m L solid-liquid ratio. The extraction process is fast,economical,time-saving,and suitable for process production.
