天然鱼肝油脂肪酸指纹图谱研究及掺假鉴定 被引量:5

Fatty acids fingerprint of natural cod-liver oil and its adulteration identification

摘要 目的建立一种天然鱼肝油脂肪酸成分的气相色谱-质谱(gas chromatography-mass spectrometry)指纹图谱分析方法,鉴定天然鱼肝油掺假问题。方法脂肪酸经柱前衍生转化为脂肪酸甲酯后,采用GC-MS联用方法测定脂肪酸指纹图谱,选取13批具有代表性的样品建立共有模式,选定15个脂肪酸峰为共有峰,棕榈酸峰为参照物峰,建立天然鱼肝油脂肪酸对照指纹图谱。借助"中药色谱指纹图谱相似度评价系统"软件,以对照指纹图谱为参比,计算样品指纹图谱相似度。结果对收集到的48批样品经指纹图谱分析和相似度评价,共检出3批掺假鱼肝油,其中2批掺假鱼油,掺假水平为20%及30%, 1批掺假植物油,掺假样品比例约为6%。结论本研究建立的脂肪酸GC-MS指纹图谱分析方法操作简便、准确可靠,结合相似度评价方法,为天然鱼肝油的质量控制和掺假鉴定提供了一种新的技术手段。 Objective To establish gas chromatography-mass spectrometry (GS-MS) fingerprint analysis method of natural cod-liver oil fatty acid composition for identification of natural cod-liver oil adulteration. Methods The GC-MS method was used to determine the fingerprint by pre-column derivatization of fatty acids into its methyl esters. A total of 13 representative samples were selected to establish a common mode, and 15 fatty acid peaks were selected as common peaks. The reference fingerprint of natural cod-liver oil was established by choosing palmitic acid peak as the referential peak. Using the “Chinese Medicine Chromatographic Fingerprint Similarity Evaluation System” software, the reference fingerprint was used as reference to evaluate the sample fingerprint similarity. Results Fingerprint analysis and similarity evaluation were performed on 48 batches of collected samples. Three batches of samples were detected as adulteration. Two of them were adulterated fish oils, and the content were 20% and 30%, respectively. The third one was adulterated vegetable oils, and the adulteration sample ratio was about 6%. Conclusion The fingerprint analysis method of the fatty acid is simple, accurate and reliable. Combined with similarity evaluation, it can provide a new technique mean for quality control and adulteration identification of natural cod-liver oil.
