摘要
目的观察镍与铬对小鼠睾丸的联合毒作用,探讨镍、铬联合作用类型及毒作用机制。方法54只昆明种雄性小鼠按3×3析因设计随机等分为9组,镍染毒剂量为0.0,2.5、5.0mg/kg·bw、铬染毒剂量为0.0、2.5、5.0 mg/kg·bw,按0.1 ml/10 g·bw连续灌胃5d,35d后处死动物,检测睾丸脏器系数、睾丸结构、睾丸细胞凋亡及细胞周期;精子数量、活力和畸型;检测睾丸组织中MDA含量、SOD、LDH、SDH活性。结果小鼠睾丸脏器系数、SOD、LDH、SDH活性随染毒剂量增加而降低,MDA含量增高(P<0.01),且有量一效关系;精子数量较对照组低(P<0.01)、精子活力下降(P<0.01)、畸形率高于对照(P<0.01),睾丸的结构受损,各染毒组小鼠睾丸细胞凋亡率和G_0/G_1期细胞百分数较对照组升高(P<0.05),各染毒组s期及G_2+M期细胞百分数较对照组低(P<0.05),并呈现一定的量一效关系,镍、铬合用的效应大于二者单独作用的效应之和(P<0.01)。结论镍和铬联合染毒可致小鼠睾丸损伤,二者的联合作用为协同毒效应;损伤机制可能与其降低睾丸细胞的抗氧化能力、抑制睾丸细胞酶活性、促进睾丸细胞凋亡等有关。
Objective To study the synergistic toxicity of nickel and chromium in testis in mice,and to explore the type of the joint toxicity and its possible mechanisms.Methods Totally,54 male Kunming mice were randomly assigned into nine groups according to 3×3 factorial design.The doses of both nickel and chromium were0,2.5 and 5.0mg/kg.bw.Nickel andchromium were administered to mice once a day at a volume of 0.1ml/10 g.bw by gavage for a consecution of five days.After being fed for 35 days.the testes were removed.The testicular organ coefficient was calculated.The changes of the testicular cell apoptosis,cell cycle and morphology were observed.The concentration of MDA and the activities of SOD,LDH and SDH in the testis were measured.In addition,epididymides were collected for determination of the sperm count? sperm motility and sperm malformation.Results When compared with the control group,the testicular organ coefficient,the activities of SOD,LDH and SDH in the testis of mice in the treatment groups were significantly decreased and MDA was increased with the increase of the exposure doses(P<0.0l).The sperm quantity and sperm motility were significantly decreased(P<0.0l),while the sperm malformation rate was markedly increased(P<0.0l).The structures of the testis were injured.The testicular cell apoptosis rate and the percentage of G_0/G_1 phase cells in all treatment groups were significantly higher than those in the eontrol group(P<0.05),while the percentage of S and G_2+M phase cells were statistically lower(P<0.05).The effects of nickel and chromium on the above indicators all exhibited obvious dose—effect relationships.The synergistic toxicity of nickel and chromium was higher than the sum of the toxicity by nickel and chromium alone(P<0.05).Conclusions Co—administration of nickel and chromium can lead to testicular damage in mice.and their ioint toxic effect is synergistic toxicity.The mechanism may involve the damage to the antioxidant enzymes in the testis.the inhibition of the activities of testicular enzymes and the acceleration ofthe testicular cell apoptosis.
