摘要
取柠条的子叶和胚轴切段作为外植体,接种于含不同激素的7种MS培养基上诱导愈伤组织发生,确定最佳愈伤组织培养基为MS+6--BA0.5mg/L,2,4--D2mg/L。在MS+6--B1mg/L,IAA10mg/L的培养基上生长的愈伤组织中,在距愈伤组织表面一定距离处常形成分生组织带,很像形成层带。分生组织带内部的细胞分化产生排列不规则的长形管状细胞,分生组织带两侧的细胞分裂产生正常的愈伤组织细胞。生长在MS+6--BA0.5/L,2,4--D2mg/L的培养基上的愈伤组织中可分化出胚性细胞(团),这样的愈伤组织转移至无激素培养基后,胚性细胞(团)可进一步分化出原胚、球形胚、心形胚、子叶胚和试管苗。
The callus was initiated in MS media with different growth-regulating substances from the cuttings of cotyledon and plunular axis of Caragana karshinkii Kom.The MS medium with 6-BA 0.5mg/L,2,4-D 2mg/L was selected as the best midium for callus culture among the seven media.The section show that the meristematic cell zone looked like cambial zone emerged in the place where have some distance from the surface of the callus growing in MS medium with 6-BA 1mg/L,IAA 10mh/L.The inner cells of the meristematic cell zone redifferented into long tubular cells,the outer (both sides)cells devided into new callus cells.The embryonic cells was redifferented in everywhere of the callus growing in the medium MS with 6-BA 0.5mg/L,2,4-D 2mg/L.When the callus was transfered to MS medium without 6-BA and 2,4-D,the embryonic cell could devided and grew into primary,ovate,corlate,cotyledonary embryods and plantlets.
