摘要
目的:建立复方制剂中苦杏仁苷HPLC定量法。方法:采用Hypersil C_(18)柱(4.6 mm×250 mm,5 μm),甲醇-乙腈-水-冰醋酸(1:1:16:0.02)为流动相,流速为0.8mL·min^(-1),检测波长210 nm,柱温40℃。以HPLC法测定了麻杏石甘汤、桂枝茯苓丸等6种自制模型制剂和橘红丸、桂枝茯苓丸胶囊等3种市售商品制剂中苦杏仁苷的含量。结果:苦杏仁苷线性范围:0.1104~1.840mg·mL^(-1)(r=0.9998);平均回收率在96.6%~103.2%;RSD在0.9%~2.6%(n=3),且操作简便、快速、适用范围广。结论:本法作为复方制剂中苦杏仁苷含量测定法是可行的。
Objective: To establish the HPLC method for the quantitative analysis of amygdalin in the traditional Chinese prescriptions. Method: In the HPLC analysis using the ODS reversed -phase column of Hypersil C18 (4. 6 mm × 250 mm, 5 μm) , the mobile phase was a combination of methanol, acetonitrile, water and glacial acetic acid in the volume ration of 1: 1: 16: 0. 02 at the flowrate of 0. 8 mL ·min-1 , and the eluate was monitored with UV absorption at 210 nm. Results: The amygdalin content in the 6 laboratory - prepared and 3 commercial traditional Chinese prescriptions containing Semen Armeniacae Amarum or Semen Persicae, such as Maxingshigan Tang, Guizhifuling Wan, etc. were determined by the HPLC method. The linear ranges of amygdalin were 0. 110 4-1. 840 mg· mL-1 with the r of 0. 999 8 and its recoveries in the 6 prescriptions were from 96. 6% to 103. 2% . The good precision was also shown by the RSD of less than 2. 6% . Conclusion: Because of a good accuracy, a-daptability and a simple procedure, the HPLC method may be useful for the chemical evaluation of the amygdalin - containing traditional Chinese prescriptions.
