福建官井洋大黄鱼AFLP指纹多态性的研究 被引量:86

<title>Genetic polymorphisms in wild and cultured large yellowcroaker Pseudosciaena crocea using AFLP fingerprinting

摘要 对福建官井洋大黄鱼(Pseudosciaenacrocea)野生种群和2个养殖群体进行5对选择性引物(共37个标本)的AFLP分析。结果表明,共检出503个不同的扩增片段(50~450bp),每对引物扩增出的片段数目为76~155个;在5对引物检出的扩增片段中,101个(20.1%)为全部37个受试个体共有,312个(62.0%)为部分野生与养殖个体共有,53个(10.5%)仅见于野生个体,37个(7.4%)仅见于养殖个体,养殖群体中出现野生种群所没有的扩增片段;野生种群、养殖1、养殖2多态片段比例分别为76.6%、70.6%、69.2%,遗传差异度分别为0.2464、0.2322、0.2299,养殖群体多态片段比例与个体间的遗传差异度均低于野生种群。养殖群体的遗传多样性水平较低,遗传变异相对贫乏。 <Abstrcat> The wild individuals of Pseudosciaena crocea were collected from Guanjing Sea area, East China Sea. The cultured individuals were collected from two cultured populations in two different culture farms in Fujian Province, which were artificially breeding F5-F7. Totally 37 individuals were used. Five primer pairs (E-AAC/M-CTC, E-AAC/M-CAA, E-ACG/MCAG, E-ACG/M-CTT, E-AGG/M-CTG) were used and 503 selectively amplified fragments were detected, ranging from 50 to 450 bp in size, and the number of fragments produced by each pair of primers were 76-155. Among the 503 fragments, 101 bands presented in the total 37 individuals, and 312 bands only presented in part of the wild fish and the cultured, 37 only in the cultured, 53 only in the wild. In the cultured individuals, some fragments presented were not found in the wild individuals. The mean percentages of polymorphic bands of the three populations varied from 69.2% to 76.6% and the mean genetic differences within populations varied from 0.229 9 to 0.246 4, and the maximums all presented in the wild population, indicating the cultured populations had lower polymorphisms. The average genetic distance between the wild and the cultured populations is larger than that between the two cultured populations.
