摘要
对红色诺卡氏菌细胞壁骨架 (N CWS)的理化性质、化学成分进行研究 ,并建立其含量测定的方法。采用化学鉴别、光谱分析及比色法。结果表明由红色诺卡氏菌P0 8菌株制备的N CWS为白色粉末 ,性质稳定 ,不溶于水、甲醇等有机溶媒。含有类脂化合物 (1 7 1 % )、多糖 (阿拉伯半乳聚糖 46 3 % )、粘肽 (主要为丙氨酸、谷氨酸、二氨基庚二酸、葡萄糖胺、胞壁酸 ) ;通过比色法测定N CWS中的阿拉伯糖并推算N CWS的含量 ,其重现性较好 ,小鼠抑瘤活性实验表明N CWS的含量与活性呈平衡关系。本文建立的测定方法可行。
The physico chemical properties, composition and the establishment of content determination method of Nocardia rubra cell wall skeleton (N CWS) were studied by chemical identification,colorimetry and spectral analyses. N CWS was a white powder, insoluble in water and organic solvent like methanol. It was composed of lipid (17 1%), polysaccharide (arabino galactan 46 3%), and mucopeptide, which was mainly composed of alanine,glutamic acid, DL α ε diaminopimelic acid, glucosamine and muramic acid. The content of N CWS was deduced from the colorimetric determination method of arabinose content, it showed that the method had a good reproducibility for routine experimental work, while the anti tumor activity was parallel to the content of the N CWS.
