摘要
目的 研究硒对体外神经细胞生长发育的直接影响及可能的作用机理。方法 采用体外新生大鼠皮层神经细胞培养法观察微量元素硒对神经细胞形态学及其表达神经元特异性烯醇化酶 (NSE)和神经生长因子(NGF)蛋白的影响。结果 中低浓度的硒能增加体外培养的神经细胞最长突起长度和细胞平均直径 ;硒能促进体外培养的神经细胞表达 NSE蛋白 ;体外培养的大鼠皮层神经细胞随着培养时间的延长 ,NGF的表达减弱 ,与对照组比较 ,硒能促进神经细胞 NGF的表达。结论 硒可能是通过促进、增强 NSE、NGF蛋白的表达 ,进而直接作用于体外培养的神经细胞 ,促进神经细胞的生长、发育与分化。
Objective To study and observe the effects of Se on the growth,development, and differentiation in cultured neurons in vitro.Methods We use the model of primary neuron culture neonatal rats' cortex in vitro, and measure the diameter of the cell and the length of the neurite, and observe the NSE and NGF expression in cultured neurons through immunohistochemical method.Results Newborn rats' cerebral neuron could grow and develop in vitro;added Se into medium could increase the diameter of the cell and the length of the neurite of the cultured neurons in vitro;added se into medium could promote NSE and NGF expression in comparison with control group.Conclusions The results indicated that selenium has an important biological significance on the neuron growth, development and differentiation in early stage in vitro.
