摘要
目的 :应用D 半乳糖制备亚急性衰老动物模型并对模型进行深入评价和探讨。方法 :2 .5月龄SD大鼠分别腹腔注射3 0 0mg·kg-1·d-1和 5 0 0mg·kg-1·d-1的D 半乳糖生理盐水溶液 5 6d ,测定动物血清中超氧化物歧化酶 (SOD)、丙二醛 (MDA)、总胆固醇和甘油三酯水平 ,解剖动物后称取胸腺、脾脏以及雌鼠卵巢的重量。结果 :与同月龄鼠对照组相比 ,高剂量雄鼠体重明显降低 (P <0 .0 1) ,SOD活性降低 (P <0 .0 5 ) ,MDA升高 ,胸腺明显萎缩 (P <0 .0 1) ,脾脏增重 (P <0 .0 5 ) ,水平接近自然衰老动物 ;高剂量雌鼠SOD活性降低 (P <0 .0 5 ) ,卵巢有 5 4.5 %出现囊肿 ,其余指标均无明显变化 (P >0 .0 5 )。结论 :应用D 半乳糖可以成功制备亚急性动物衰老模型 ,而雄性大鼠相对于雌性更为敏感 ,因此 ,首选雄性SD大鼠制备动物模型用于衰老试验研究。
Objective The development and estimation of an animal subacute aging model induced by D galactose were reported in the paper.Methods The experiment was conducted on the 2.5 month old SD rats injected with D galactose 300?mg·kg -1 ·d -1 or 500?mg·kg -1 ·d -1 for 56 days by intra peritoneum.The level of SOD,MDA,CH and TG were measured and the weights of rats thymus,spleen and ovaries of female rats was weighed.Results The weights of male rats in group 500?mg· kg -1 ·d -1 were lower than that of the rats of control group without D galactose ( P <0.01),and the serum SOD activity was lower too( P <0.05),but the content of serum MDA were higher.And the result indicated that the ratio of rats thymus weight with body weight was reduced ( P <0.01) and that of spleen weight with body weight was increased in group 500?mg·kg -1 ·d -1 compared with control group.There were no significant differences between the indexes of female rats that were given D galactose 500?mg·kg -1 ·d -1 and the female rats in control group expect that the serum SOD activity was higher than the latter ( P <0.05).And 54.5% of the female rats that were given D galactose 500?mg·kg -1 ·d -1 had ovaries cyst.Conclusion Our results show that the rat model of subacute aging can be established by intra peritoneum injection of D galactic and the male rats are the first choice animals.
