摘要
本研究对不同性别和不同年龄组的小鼠颌下腺颗粒曲管(GCT)进行了电镜观察,并对其产生神经生长因子(NGF)的功能进行了免疫组化和形态计量研究,结果显示:颗粒曲管的截面积、周长、等效直径及其NGF的相对含量均与年龄呈明显的正相关,在同年龄组中雄性小鼠的上述参数明显大于雌性小鼠。电镜观察可见:2周龄的颌下腺中开始出现GCT,但细胞内的颗粒尚不明显;1月龄的GCT细胞内的颗粒明显可见,且随年龄的增长而逐渐增多、增大。分泌颗粒的大小不等,电子密度也不尽一致。3月龄小鼠的GCT细胞中分泌颗粒占据了细胞的上2/3区域。管腔内充满了分泌物。实验结果为评估颌下腺的发育状况和生理功能,为确立从颌下腺提取NGF的最佳时间提供了重要的实验依据。
In this study granular convoluted tubules in the submandibular glands of mice with different sexes and in different age groups were examined under the electron microscope and their function of producing neural growth factor (NGF) was studied immunohis to chemically and mor--phometrically. The results showed that the sectional area, circumference, equivalent diameter and the relative content of NGF of the granular convoluted tubules were positively related with their ages and in the same age group the above-mentioned parameters in the male mice were greater than those in the female. It was observed under the electron microscope that GCT began to appear in the submandibular glands of mice of two weeks-age but the granules in the cells were not evident; the granules in the GCT cells of mice of one month-age were apparently seen and their numbers and size were gradually increased with their ages. The secreted granules varied in size and their electronic densities were different. The secreted granules in the GCT cells in mice of three months-age occupied the upper 2/3 area and the cavity of the tubules was full of secretions. The results of this experiment provides the important experimental basis both for assessing the development and physiological function of the submandibular gland and for determining the best time to collect NGF from the submandibular gland.
