摘要
为解决青钱柳资源稀缺问题以及为采用生物反应器规模化生产青钱柳多糖,采用单因素和正交试验研究了几种大量营养素对青钱柳悬浮细胞生长及多糖质量分数的影响。单因素试验结果表明:添加40g/L蔗糖有利于悬浮细胞的生长,而50g/L蔗糖时多糖质量分数最高;NO3-:NH4+为50︰10、总氮30mmol/L、磷酸盐1.25mmol/L、Mg2+2mmol/L时细胞干重和多糖质量分数均最高;4mmol/L Ca2+适合细胞的生长,但2mmol/L Ca2+更有利于多糖的合成。正交试验结果表明:利于细胞生长的大量营养素组合为40g/L蔗糖+30mmol/L总氮+2mmol/LMg2++4mmol/L Ca2+;而适于细胞多糖含量提高的组合为40g/L的蔗糖+30mmol/L总氮+3mmol/L Mg2++3mmol/L Ca2+。
In order to provide reference for production of Cyclocarya paliurus polysccharide on a large scale in bioreactor,and solve the problem of Cyclocarya paliurus resources scarcity,the effects of nutrition conditions on on the growth and polysaccharide content of C.paliurus suspension culture cells were studied in this manuscript.The single factor experiment and orthogonal test were applied to optimize the nutrition conditions and the result were listed as follows: for cell growth: 40 g/L sucrose+30 mmol/L total nitrogen +2 mmol/L Mg2++4 mmol/L Ca2+.for polysaccharide production:40 g/L sucrose+30 mmol/L total nitrogen +3 mmol/L Mg2++3 mmol/L Ca2+.
