摘要
目的 建立血及组织中美洛昔康药物浓度的高效液相色谱 (HPLC)检测法。方法 采用高效液相色谱法联用紫外检测器 ,检测血及组织中美洛昔康药物浓度。流动相∶甲醇∶水∶冰乙酸∶乙腈 (6 0 0∶5 0 0∶2 0∶5 0 )V/V ;检测波长 :35 5nm。结果 血及组织中美洛昔康最低检测浓度 0 0 39mg·L-1,检测限 1 95ng ,Mel在血及组织中能得到很好的分离 ,线性范围为 0 0 39~ 10mg·L-1,回归方程 :Y =3132 81 9X +4 414 3,r =0 9998,P <0 0 5 ,回收率在 85 6 7%~ 95 0 9%之间 ,日间、日内的变异均在5 6 %以下 ,足爪局部回收率在 80 13%~ 94 5 6 %,符合生物样品分析的要求。结论 本法操作简便 ,灵敏度高 ,准确性好。
AIM To set up a high performance liquid chromatography(HPLC)method for determine the concentration of Meloxicam in serum and local tissue. METHODS A high performance liquid chromatography(HPLC)method with ultraviolet detection was used.The mobile phase consisted of methanol∶Water∶Acetic acid∶Acetonitrile=600∶500∶20∶50 with a flow rate of 1.0 ml·min -1,and the UV absorbance detector was set at 355 nm. RESULTS Mel in serum and local tissue could be well separated.The limit of detection was 1.95 ng.A good linearity of Mel was obtained from 0.039 mg·L -1 to 10 mg·L -1 with r= 0.999 8 (P<0.05).The recovery in serum was between 85.67% and 95.09%,and the recovery in local tissue was between 80.13% and 94.56%.All of these showed the method fitted for analytic requirement of Mel preparation.CONCLUSION The method is simple,accurate and sensitive.
