摘要
从水稻植株上分离到 1株对水稻纹枯病菌有较好拮抗作用编号为 B34的菌株 ,室内对水稻纹枯病菌的抑菌带达到 10 .3mm。经生理、生化检测及细胞壁化学成分分析 ,确定其为致黄假单胞菌 (Pseudomonas aureofaciens)。其代谢粗提物经蛋白酶作用 ,拮抗作用消失。经 70 %饱和度的 (NH4) 2 SO4沉淀 ,透析后得到粗杀菌蛋白 ,分别上样 Sephadex G- 10 0和DEAE-纤维素 - 32 L KB柱层析系统 ,分离纯化了杀菌蛋白 ,经 SDS- PAGE检测其分子量约为 33.0 k Da,经低温、真空干燥 。
A strain B34 against Rhizoctonia solani was screened from rice plant,in vitro the inhibition zone width was 10 3 mm. Results of analysis on chemical components in cell wall, physiological and biochemical characters of B34, indicated it was Pseudomonas aureofaciens. When its crude extracts were treated by proteinase K and trypsin, the antagonism disappeared. The crude protein of B34 was obtained with 70% staturated (NH 4) 2SO 4. The protein was purified by ion exchange and gel filtration LKB chromatography. One antagonistic peak was attained and showed only single band with 33.0 kDa by SDS PAGE. The purified protein crystal of antiseptic protein was obtained.
