摘要 目的:本实验意在确立培养的兔视网膜Muller细胞上腺苷/腺苷酸受体的存在。方法:取材于兔视网膜组织,完成Muller细胞的培养;而后应用自由钙指示剂Fura-2AM,及ARGIS-100/CA系统,在受体拮抗剂有或无时,加入ATP或其同系物,监测细胞内自由钙的变化,从而确定受体的存在。结果:100nm ATP可引起Muller细胞内钙离子的变化,A1、A2、P2x、P2y亚型受体激动剂在不同的浓度下也各自具有类似的作用,ATP及其同系物的这一作用不能被电压依赖性钙通道阻滞剂Nifedipine所抑制,A1、A2亚型受体拮抗剂可分别部分地抑制ATP的作用,但完全抑制亚型受体激动剂的作用。结论:培养的兔视网膜Muller细胞上具有腺苷/腺苷酸受体(包括A1、A2、P2x、P2y亚型受体)。 To determine whether there are adenosine (A1, A2)/purinergic (P2x, P2y) receptors on cultured rabbit Muller cells. Methods. Muller cells were cultured from adult rabbit retina. Intracel-lular calcium concentration was minotered by a videospetrofluorome try (ARGUS-100/CA) using Fura-2AM, a calcium indicator. Calcium transient after administration of ATP or its analogues was observed with or without receptor antagoist. Results: ATP at 100nM can induce calcium transient in most cells.Al agonist, A2, P2x, P2y receptor agonists at diffirent concentrations can increase the cytosolic calcium in some Muller cells, and is not blocked by Nifedipine-a voltage dependent calcium channaJ blocker. Al, A2 receptor antagonist can partially block the calcium increase caused by ATP, but can completely block the calcium changes induced by Al, A2 receptor agonist. Conclusion. Cultured rabbit Muller cell have Al, A2, P2x, P2y receptors.
