摘要
利用双微电极电压钳制技术研究丹参注射液(以下简称丹参)对离体家兔浦肯野纤维慢内向电流(Isi)的电生理作用。结果表明,丹参在浓度为1.0mg/ml 和2.0mg/ml,灌流20min 时,能明显抑制 Isi 峰值,由给药前的(76.25±20.83)nA 降至(53.13±18.70)nA 和(36.00±10.84)nA(P<0.05,P<0.01),对 Isi 峰值抑制率分别为31%和53%,呈浓度依赖性抑制作用;在浓度为0.5mg/ml,刺激频率为1.0Hz,丹参在开始抑制 Isi 的峰值不明显,随着刺激数目增加,以后抑制逐渐加强,直至稳态,呈使用依赖性阻滞作用。以上表明丹参具有钙拮抗剂作用。
Two-microelectrode voltage clamp technique was used to study the effects of Salvia miltiorrhiza(SM)on slow inward cur- rents(Isi)d isolated rabbit cardioc-Purkinje fibres.The results showed that SM at 1.0 mg/ml and 2.0 mg/ml inhibited Isi from 76.25±20.83 nA to 53.13±18.70 nA and 36.00±10.84 nA respectively(P<0.05 and 0.01)after 20 min perfusion.SM inhibited amplitude of Isi were 31% and 53% respectively.It showed that concentration-dependence relationship.At 0.5 mg/ ml and a frequency of 1.0 Hz,SM inhibited Isi uncertainly,as the frequency increased,the inhibited Isi become evidently.It showed that use-dependence blocked Isi.It was concluded that SM had function of calcium channel antagonist.
