摘要
【目的】测定西藏的普通栽培天麻、有机肥栽培天麻与野生天麻中天麻素和柠檬酸的含量。【方法】采用RP-HPLC法。色谱柱Hypersil C18(4.6 mm×250 mm,5μm);柱温:25℃;检测波长分别为:220 nm和214 nm;流动相分别为:乙腈-0.05%磷酸溶液(3∶97)和0.8%NH4H2PO4水溶液。对西藏的普通栽培天麻、有机肥栽培天麻与野生天麻中的天麻素和柠檬酸进行含量测定。【结果】不同栽培方式得到的天麻中天麻素含量分别为:0.12%,0.14%和0.24%;柠檬酸的含量分别为:0.0234%,0.0298%和0.0266%:野生天麻中天麻素的平均回收率为98.20%(n=6);柠檬酸的平均回收率为95.49%(n=5)。【结论】不同的栽培方式对天麻中天麻素和柠檬酸的含量影响较大,其中野生天麻样品中天麻素含量最高,柠檬酸的含量仅次于普通栽培天麻。
Objective To determine the contents of gastrodin and citric acid from different cultivation of gastrodia elata in Tibet by HPLC. Methods High-performance liquid chromatography (HPLC) was conducted on Hypersil C18 (4.6 mm×250 mm, 5 μm) with a mobile phase of acetonitrile-0.05% phosphoric acid (3 : 97) and 0.8%NH4H2PO4 solution at a detection wavelength of 220 nm and 214 nm respectively. Results The content of gastrodin from different cultivation of gastrodia elata were 0.12%,0.14% and 0.24%, and the content of citric acid were 0.0234%,0.0298% and 0.0266%.The average recovery rate of gastrodin from nature was 98.20% (n=6), and the content of citric acid was 95.49% (n=5).Conclusion There are significant differences in the content of gastrodin and citric acid between different cultivation of gastrodia elata in Tibet.
