摘要
目的:观察樟芝升白汤对环磷酰胺所致免疫力低下小鼠模型的肝肾毒性作用,为樟芝升白汤的临床用药安全提供参考依据。方法:将KM小鼠48只随机分为空白组,模型组,给药(等效剂量)组,模型给药组(低、中、高)6个组。给药14d后,眼眶静脉丛取血后处死小鼠,全自动生化分析仪检测血清ALT、AST、GGT、BUN、CREA、UA的含量;取肝脏、肾脏,计算其脏器系数;常规病理学切片,HE染色观察肝脏、肾脏组织形态学变化。结果:与空白组比较,给药组和模型给药中、高剂量组小鼠体重增长无差异(P>0.05),肝肾功能指标水平无差异(P>0.05),模型组及模型给药低剂量组体重增长有差异(P<0.05),肝、肾功能指标水平(ALT、AST、GGT、CREA、UA)均有差异(P<0.05);空白组、模型组、给药组与模型给药(低、中、高)组肝脏、肾脏系数比较无差异(P>0.05),但与药物浓度成负相关关系;病理改变结果:模型组和模型给药低剂量组肝肾组织均损伤严重,模型给药中、高剂量组及给药组肝肾组织无损伤。结论:樟芝升白汤对环磷酰胺所致免疫力低下小鼠模型无肝肾毒性作用,并有一定保护作用。
Objective: To observe effects of hepatotoxicity and nephrotoxicity of the Zhangzhi Shengbai decoction on immunocompromisedfrom cyclophosphamide in model rats, in order to provide a reference for clinical medication safety. Methods: 48 rats were randomly dividedinto the blank group, the model group, the treatment group (equivalent dose), the model treatment groups (low, middle, high dose). In 14 days ofadministration, orbital venous plexus blood was taken when the mice were executeed; automatic biochemical analyzer was used to measure theserum ALT, AST, GGT, BUN, CREA, UA levels. The morphological change of liver and kidney tissue was observed under light microscope.Results: Compared with the blank group, differences of weight in the model group and the model treatment of low dose group were significant(P<0.05); and liver and kidney function indexes (ALT, AST, GGT, CREA, UA) in the model group and the model treatment of low dose groupshowed significant differences (P<0.05). The coefficients of liver and kidney among six groups showed no significant difference (P>0.05), andnegatively correlated with drug concentration. Pathology results: Liver and kidney tissues in the model group and the model treatment of low dosegroup were damaged severely. Conclusion: The Zhangzhi Shengbai decoction on immunocompromised from cyclophosphamide in model ratsshowed no toxicity to the liver and kidney, with a certain protective effect.
