摘要
通过紫外诱变的方法研究不同时间紫外诱变对平菇菌丝体生长量、干物质重量、可溶性蛋白质含量,以及其对菌丝体生长过程中产生的代谢酶活力的影响,结果表明:当紫外诱变时间达到5min时,平菇长势最好,干物质重量达2.01g,而且菌丝体中的可溶性蛋白含量最高(3.09μg/g)。与此同时菌丝体在代谢过程中产生的纤维素酶和超氧化物歧化酶(SOD酶)活力均有显著性提高,与对照相比诱变后纤维素酶的活力提高1.887倍,SOD酶的活力提高了1.29倍。在此基础上进行的遗传稳定性试验表明:平菇菌丝体经过5min诱变后纤维素酶的活力达1.7~1.9倍,SOD酶活力提高了1.2~1.5倍。得出平菇菌丝体经紫外诱变后能够育选出长势更旺,代谢酶活力更强的平菇菌种,为平菇优良菌种的选育奠定了理论基础。
This paper focuses on the effect of different period of UV mutation time (1,3,5,7,9 min) onthe growth and metabolism of P. ostreatus mycelium. The results showed that when the UV mutation timereached at 5 min,not only the mycelium’s growth vigor was the best and the dry,weight could achieve 2.01gbut also the content of soluble protein of mycelium was the highest (3.09μg/g). What’s more,the cellulase andsuperoxide dismutase (SOD) were improved significantly during the metabolic process. Compared with control,after 5min UV radiation the cellulase activity and superoxide dismutase increased to 1.887 and 1.29 times,respectively. The genetic stability test indicated that,after 5 min ultraviolet mutation,both of cellulase activityand superoxide dismutase activity were increased to 1.7~1.9 and 1.2~1.5 times,respectively.The resultsproposed that UV mutation breeding technology could select better mycelium,which growth vigor was strongerand metabolic enzymes’activity was better than the one that was without UV radiation. It could provide atheoretical principle for future study on P. ostreatus.
