摘要
采用种子浸渍法分析秋水仙素对小桐子(Jatropha curcas)的诱变效果,通过系统比较不同处理下发芽势、发芽率、植株形态、气孔大小、保卫细胞大小和气孔密度等的变化,对加倍效果进行初步筛选与鉴定。实验结果表明:和对照组(无菌水浸渍)相比,不同处理对小桐子种子的发芽势无显著影响,但对发芽率表现出了抑制作用且差异显著;在一定范围内随着处理时间的延长发芽率先升高后降低;0.5%秋水仙素处理24 h时胚根膨大率最高(64%);秋水仙素诱导后植株气孔变大、保卫细胞变大、气孔密度显著减小,具有多倍体特征。
The mutagenic effect of colchicine onJatropha curcas was analyzed by seed dipping method.The germination potential,germination rate,plant morphology,pore size,guard cell size and stomata density after different treaments were systematically compared for the original screening and identification of the doubling effect.The results showed that compared with the control group,experimental groups had no obvious influence on the germination potential ofJatropha curcas seeds,but there was significant inhibition of the germination rate,and the germination rate firstly increased and finally decreased with the extension of time in a specific range.The highest radicle expansion ratio was64%after the treatment of0.5%colchicine for24h.The plants induced by colchicine showed obvious characteristics of polyploid which guard cells and pore sizes of plants became large,and stomata density decreased significantly.
