ASIC3在实验性大鼠Ⅲ型前列腺炎模型慢性骨盆疼痛中的表达

Significance of The Expression of ASIC3 in Chronic Pelvic Pain of Experimental Rat Model of Type Ⅲ Prostatitis

摘要 目的探讨三型酸敏感离子通道(ASIC3)与Ⅲ型前列腺炎(CPPS/CP)慢性骨盆疼痛的关系.方法选取雄性SD大鼠45只,分为对照组、Ⅲ型前列腺炎模型/EAP组和干预组,每组15只.建立EAP模型过程中,予Von-Frey测痛纤维于第5、10、20、30、40 d于对照组和EAP组进行骨盆区域痛觉测试.建模成功后,干预组予阿米洛利连续腹腔内注射10 d,每2 d对其及EAP组进行痛觉测试.选取3组大鼠的前列腺,进行HE染色染色病理学检查;并通过免疫组化和Western blot方法检测ASIC3的表达.结果 Von-Frey测痛结果显示:EAP模型组大鼠骨盆疼痛表现较对照组明显,干预组大鼠骨盆疼痛表现较EAP组减轻.HE染色提示:EAP组大鼠前列腺间质内淋巴细胞和中性粒细胞浸润,腺体周围充血,腺腔不规则改变.对照组未见明显炎症细胞浸润.与EAP组对比,干预组腺体充血及炎症细胞浸润情况无明显改变.免疫组化染色提示:EAP组大鼠的前列腺组织中,ASIC3表达较对照组表达增加(主要表达于细胞核与细胞浆中)(P<0.01),干预组较EAP组减少(P<0.05).Western blot提示:EAP组大鼠的前列腺组织中,ASIC3较对照组表达增加(P<0.01),干预组较EAP组减少(P<0.01).结论 EAP大鼠前列腺组织中ASIC3表达增加,ASIC3是导致Ⅲ型前列腺炎骨盆区域疼痛介质之一,可作为干预及治疗Ⅲ型前列腺炎的其中一个靶向指标. Objective To explore the relationship of type3of acid-sensitive ion channels(ASIC3)andthe chronic pelvic pain of typeⅢprostatitis(CPPS/CP).Methods Randomly allocation was taken to averagelydivide45SD rats into three groups:Control group(intraperitoneal injection of0.9%sodium chloride),Experimental Autoimmune Prostatitis model group/EAP group(intraperitoneal injection of the mixed suspension ofComplete Freund's adjuvant and prostate tissue),Intervention group(EAP rats treated with amiloride).Tactileallodynia was quantified using Von-Frey filaments as a measure of pelvic pain behavior.And Von-Frey filaments wasperformed on the5th,10th,20th,30th and40th day in control group and EAP group.When the EAP model wasestablished successfully,the rats in intervention group were given drugs for10days,and during this time we testedthe tactile allodynia in intervention group and EAP group every other days.Mouse prostate were collected from miceand were processed for H&E staining,while the expressions of mast cell tryptase were measured byimmunohistochemistry and Western blot.Results Von-Frey test results showed that the pelvis pain was significantlymore obvious in EAP model group than that in control group.However,the pelvis pain in EAP model group waslighter than that in EAP group.HE staining showed that the lymphocytes and neutrophils infiltrated in the prostate of EAP rats,and the surrounding glands were congested.No inflammatory cell was founded in the control group.Thecongestion of the gland and the infiltration of the inflammatory cells were not alleviated,when intervention groupcompared with EAP group.Immunohistochemical staining results showed that the expression of ASIC3wassignificantly increased(P<0.01)in EAP group when compared with control group,while in intervention groupshown reduced expression(P<0.05).Western blot results showed that ASIC3expressed higher in EAP group whencompared with control group(P<0.01),while in intervention group shown reduced expression.ConclusionsThese results indicate that the number of mast cells and degranulated mast cells in prostate of EAP rats aresignificantly increased.ASIC3are one of the most important mediators of chronic pelvic pain caused by typeⅢprostatitis,which can be used as a target for therapeutic intervention and treatment in typeⅢprostatitis.
