摘要
目的探讨阿莫西林克拉维酸钾最低杀菌浓度体外诱导细菌耐药试验研究。方法将金黄色葡萄球菌标准菌株ATCC29213作为研究对象,将阿莫西林克拉维酸钾作为抗菌药物,检测方法是以微量稀释法来定量测定经过特定诱导天数时的MBC值,用1/2MBC浓度来对金黄色葡萄球菌进行多步体外诱导试验共34天,并对试验结果金黄色葡萄球菌株MBC的变化进行记录,使用全自动微生物药敏鉴定仪来分析诱导后金黄色葡萄球菌株进行阿莫西林克拉维酸钾耐药性试验结果。结果金黄色葡萄球菌标准菌株的MBC值经过阿莫西林克拉维酸钾体外诱导试验,在第9天时MAC值明显升高,在经过34天诱导后MBC值是标准菌株MBC值的32倍,Pearson相关系数显示亚MBC抗菌药物时间和标准菌株的耐药性呈正相关性(r=0.715,P<0.001)。诱导出的金黄色葡萄球菌株对阿莫西林克拉维酸钾药物100%耐药。结论亚MBC阿莫西林克拉维酸钾能够体外诱导金黄色葡萄球菌耐药菌株,其耐药性跟诱导时间成正比。
Objective To investigate the antibiotic resistance of amoxicillin and clavulanate potassium in vitro.Methods The standard strains of Staphylococcus aureus ATCC29213as the research object,the amoxicillin and clavulanate potassium as antimicrobial agents,the detection method is determined by the specific induction of days when the value of MBC by micro dilution method to quantitative,with the concentration of1/2MBC for multi step test for34days in vitro of Staphylococcus aureus,and change the test results of Staphylococcus aureus strain MBC were recorded to analyze the induction of Staphylococcus aureus strains of amoxicillin and clavulanate potassium resistance test results using automated microbial susceptibility testing instrument.Results The standard strains of Staphylococcus aureus MBC value after amoxicillin and clavulanate potassium in vitro test in9days when the MAC value increased significantly,after34days of induction when the MBC value is32times the standard strain of MBC,Pearson correlation coefficient showed a positive correlation between sub-MBC antimicrobial drug time and drug resistance(r=0.715,P<0.001),induced by Staphylococcus aureus strains of amoxicillin and clavulanate potassium100%drug resistance.Conclusion MBC Amoxicillin clavulanate can induce Staphylococcus aureus resistant strains in vitro,and its drug resistance is proportional to the induction time.
