摘要
目的:建立HPLC-DAD法测定一枝蒿中一枝蒿酮酸、绿原酸、木犀草素在不同生长期的含量。方法:色谱柱为Agilent ZORBAX SB-C_(18)(4.6 mm×250 mm,5μm),流动相为0.5%冰乙酸(A)-乙腈(B),梯度洗脱(0~10 min,10.5%~14.5%B;12~23 min,22%~26%B;29~46 min,27.5%~28.5%B),检测波长一枝蒿酮酸250 nm、绿原酸和木犀草素338 nm,柱温35℃,流速:1 mL/min。结果:一枝蒿酮酸0.306 3~4.90μg(r=0.999 9);绿原酸在0.1338~2.14μg(r=0.999 4);木犀草素在0.023 9~0.143μg(r=0.999 1)线性范围内线性关系良好。平均回收率分别为95.7%(RSD=0.75%),103.2%(RSD=1.77%),100.7%(RSD=2.64%)。结论:该方法简便,可靠,准确度高,能够有效监测一枝蒿中3种化学成分在不同生长期的含量变化,为确定合理的采收时间提供理论依据。
Objective:To establish the HPLC-DAD method to determinate the content of Rupestonic acid,Chlorogenic acid,and Luteolin in Artemisia Rupestris L.at different growth stages.Methods:The chromatographic column was Agilent ZORBAX SB-C18(4.6mm×250mm,5μm)with0.5%glacial acetic acid(A)-acetonitrile(B),gradient elution(0~10min,10.5%~14.5%B;12~23min,22%~26%;29~46min,27.5%~28.5%B).The detection wave of Rupestonic acid was about250nm,and Chlorogenic acid and Luteolin was about338nm.The column temperature was35℃,and the flow rate was1mL/min.Results:The linearity ranges of Rupestonic acid,Chlorogenic acid and Luteolin were0.3063~4.90μg(r=0.9999),0.1338~2.14μg(r=0.9994),0.0239~0.143μg(r=0.9991),which showed that there was a good linear relationship.The average recoveries were95.7%(RSD=075%),103.2%(RSD=1.77%),and100.7%(RSD=2.64%)respectively.Conclusion:This method is simple,reliable and accurate.It can effectively monitor the changes of the three chemical components in Artemisia Rupestris L.at different growth stages,and provide the theoretical basis for determining the reasonable harvest time.
