摘要
为给不同蛋白酶家族的鉴定及蛋白酶抑制剂的筛选提供参考,以甘蓝型油菜鼎油杂4号为试验材料,建立基于Gelatin-SDS-PAGE的油菜蛋白酶的酶谱检测方法,并对不同温度和pH对油菜叶片中蛋白酶酶谱的影响进行对比。结果表明:甘蓝型油菜叶片蛋白酶在37℃、pH 7.6时,凝胶酶谱中酶解底物被蛋白酶消化后呈现的条带透明度较强,相应区域存在蛋白酶且其活性较强。甘蓝型油菜中叶片蛋白酶主要分布在2个区域,区域1蛋白酶的相对分子质量在180kDa以上,区域2蛋白酶的相对分子质量介于63~100kDa。
The protease zymogram in leaves of Dingyouza 4,a Brassica napus variety,is detected at different temperature and pH by the established rape protease zymogram detection method based on Gelatin-SDS-PAGE to provide a reference for identification of different protease family and screening of protease inhibitor.Results:The band from the enzymolysis substrate in gelatin zymography digested by protease shows a strong transparency and there is protease with a high activity in the corresponding area when the protease in B.napus leaves is at 37℃ and pH7.6.The protease in B.napus leaves distributes in two areas mainly.The relative molecular mass of protease in Area I and Area II is above 180 kDa and 63~100 kDa respectively.
