摘要 目的:建立同时测定鼠曲草不同部位3种黄酮类化合物(槲皮素、木犀草素、芹菜素)含量的高效液相色谱法。方法:采用Diamonsil-C18(4.6 mm×250 mm,5μm)色谱柱,流动相:甲醇-0.2%磷酸(50∶50),等度洗脱,流速:1 m L/min,检测波长:360 nm。结果:槲皮素、木犀草素、芹菜素的进样量分别在0.122~2.434、0.147~2.940、0.094~1.880μg范围内与各自峰面积呈良好线性关系,r分别为0.999 4、0.999 7、0.999 6,方法的精密度、重复性均满足分析要求,方法的回收率均大于98%。结论:该方法简单、快速、高效,可用于鼠曲草不同部位中3种黄酮类化合物的含量测定。 Objective To establish a high performance liquid chromatography(HPLC)method for simultaneous determination of three flavonoids(quercetin,luteolin,apigenin)in different parts of Gnaphalium affine D.Don.Methods Diamonsil-C 18(4.6 mm×250 mm,5μm)column was adopted.The mobile phase was methanol-0.2%phosphoric acid(50∶50)solution with the isocratic elution at the flow rate of 1.0 mL/min.The column temperature was 30℃and the detection wavelength was 360 nm.Results There was a good linear relationship between content and peak area when the quercetin,luteolin and apigenin were within the range of 0.122~2.434(r=0.9994),0.147~2.940(r=0.9997),0.094~1.880μg(r=0.9996).The performance characteristics of precision and reproducibility of the developed HPLC method fulfilled the analysis requirements.Conclution The method is simple,rapid and efficient,and applicable for determination of three flavonoids in different parts of Gnaphalium affine.
