摘要
目的探究氨基酰化酶-1(ACY-1)在肝细胞癌(肝癌,HCC)组织中的表达及其临床意义。方法使用实时荧光定量PCR(qRT-PCR)技术对HCC患者的癌组织和相应癌旁组织中的ACY-1 mRNA表达进行检测;采用蛋白质印迹法(Western blot)检测肝癌和癌旁标本中ACY-1蛋白的表达,同时结合免疫组化的方法分析了ACY-1蛋白的表达与HCC的关系。结果 qRT-PCR和Western blot结果表明癌旁组织中ACY-1的mRNA和蛋白的表达高于肝癌组织。免疫组化结果与Western blot结果一致。另外,通过对ACY-1免疫组化结果与临床病理参数分析发现,ACY-1蛋白表达与AFP水平密切相关。结论 ACY-1可能与肝癌的发生发展密切相关。
Objective To investigate the expression of aminoacylase 1(ACY-1)in hepatocellular carcinoma(HCC),and its clinical significance.Methods The expression of ACY-1 mRNA was detected by using qRT-PCR in HCC tissues and the corresponding adjacent non-HCC tissues.Western blot was used to detect the ACY-1 expression of HCC tissues and the corresponding adjacent tissues.Meanwhile,Immunohistochemical staining was used to analyse the relationship between expression of ACY-1 and HCC.Results qRT-PCR and Western blot showed that ACY-1 mRNA and protein expressed lower in matched tissues.Immunohistochemical staining also showed the same expression trend of ACY-1.Moreover,the relationship between ACY-1 protein expression level and clinical pathological parameters was analyzed by using IHC,which showed that the expression of ACY-1 protein was correlated with the serum alpha fetoprotein(AFP)level.Conclusion ACY-1 might be associated with the progression of HCC.
