摘要
在病毒密度梯度离心纯化过程中,利用光对不同大小粒子发生散射的强弱,即丁达尔效应,定位超速离心后形成的离心区带;以伪狂犬病毒为研究对象,以不连续梯度蔗糖为介质,超速离心后,通过光束对离心区带生的丁达尔效应进行精确分层取样,并对处理后的离心区带样品分别进行透射电镜观察,发现区带3中存在大量符合伪狂犬病病毒形态学特征的病毒颗粒。本研究将光学物理现象与经典病毒纯化方法相结合,大大提高了病毒纯化成功率,对密度梯度离心法提纯各种病毒或者蛋白的实验操作均有指导意义。
During the process of virus density gradient ultracentrifugation purification,the centrifugal zone formed after ultracentrifugation can be located by the scattering intensity of light of different-size particles,which was also known as the Tyndall effect.In this article,taking Pseudorabies virus(PRV)as the research object and discontinuous gradient sucrose as the medium,the particles in centrifugal zone was sampled accurately and hierarchically after ultracentrifugation,then the obtained samples were observed by electron microscope respectively.According to the observation,a large number of virus particles conforming to the morphological characteristics of PRV were found in the third layer.In this study,the optical-physical phenomena was combined with the classical virus purification method,which greatly improved the success rate of virus purification and had great significance for the purification of various viruses and proteins using density gradient ultracentrifugation.
