芦荟大黄素对宫颈癌HeLa细胞生物学行为的影响 被引量:4

Effects of Aloe emodin on cell biological behaviors of human cervical carcinoma Hela cell in vitro

摘要 目的探讨芦荟大黄素(Aloe emodin,AE)对宫颈癌He La细胞生物学行为的影响。方法宫颈癌He La细胞经过培养后,设立实验组(分别采用0、20、40、80、160μmol/L浓度AE进行处理),另设对照组(以等体积的DMSO溶液代替药物)。处理后分别检测0、24、48、72 h He La细胞增殖情况;分析培养48 h后细胞周期、细胞凋亡、细胞迁移和侵袭能力的变化;检测Caspase-3和Bcl-2蛋白的表达。结果与对照组比较,10μmol/LAE在处理24 h时对Hela细胞增殖抑制作用不明显,但当时间延长后抑制作用逐渐增大,浓度>20μmol/L时抑制作用比较明显,与对照组比较差异有统计学意义(P<0.05),AE对宫颈癌Hela细胞作用48 h的IC50为58.3μmol/L。AE处理后48 h后,除外10μmol/L组,其余浓度实验组宫颈癌Hela细胞周期的G2/M期比例较对照组均明显增加,迁移及侵袭实验中的穿模细胞数较对照组均明显减少,Hela细胞中Caspase-3表达较对照组显著增加,Bcl-2蛋白表达较对照组显著减少,差异均有统计学意义(P<0.05),实验组出现凋亡现象,且随AE浓度增加,凋亡率也增加,与对照组比较,差异均有统计学意义(P<0.05)。结论 AE能通过阻滞细胞周期于G2/M期而抑制宫颈癌He La细胞的增殖,促进细胞凋亡,降低细胞的迁移及侵袭能力,其作用机制可能与Caspase-3表达上调,Bcl-2蛋白表达下调有关。 Objective To investigate the effects of Aloe emodin(AE)on cell biological behaviors of cervical carcinoma Hela cell in vitro.Methods The cervical carcinoma Hela cells were cultured in vitro and treated with AE at the concentrations of 0,20,40,80,160μmol/L,respectively(experimental groups),moreover,the cells that were treated with the same volume DMSO instead of AE were served as control group.The cell proliferation state,cell cycle,cell apoptosis,cell migration and invasion abilities as well as the expression levels of Caspase-3 and Bcl-2 protein were observed and compared between two groups.Results As compared with those in control group,the inhibition effects of AE(10μmol/L)on cell proliferation were not obvious after 24 hour,however,with the time going on,the inhibition effects were gradually increased,and the inhibition effects of AE(20μmol/L)were significant(P<0.05).The IC50 of AE effects on Hela cells after 48 hours was 58.3μmol/L.On 48 hours after the cells were treated with AE,except for 10μmol/L concentration group,the cell proportion at G2/M phase of cell cycle in the other concentration groups was significantly increased,as compared with that in control group,however,the cell migration ability and invasion ability were significantly decreased(P<0.05).Moreover the expression levels of Caspase-3 protein were obviously increased,however,the expression levels of Bcl-2 protein were significantly decreased,as compared with those in control group(P<0.05).Besides cell apoptosis was found in experimental groups,with a dose-dependent manner,as compared with that in control group(P<0.05).Conclusion AE can inhibit the proliferation of cervical carcinoma Hela cells by blocking cell cycle at G2/M phase,inducing cell apoptosis,decreasing cells’migration and invasion abilities.Moreover its action mechanism may be correlated with the upregulation of Caspase-3 protein and downregulation of Bcl-2 protein.
