摘要
目的探讨中国新疆哈萨克族人群红细胞MNS、Duffy、Kell、Dombrock、Diego、Kidd、Scianna、Colton和Lutheran 9种血型系统基因频率分布情况,为人类群体遗传学、临床用血及稀有血型库建设等提供依据。方法采用PCR-SSP法对196例新疆哈萨克族人群9个血型系统进行基因分型和统计学处理,并与其他人群进行比较。结果 (1)获得中国新疆哈萨克族人群9种稀有血型系统等位基因频率,依次为:M=0.645 4,N=0.354 6,S=0.142 8,s=0.846 9,Fy^a=0.757 6,Fy^b=0.242 3,K1=0.002 6,K2=0.997 5,Do^a=0.221 9,Do^b=0.778 0,Di^a=0.035 7,Di^b=0.964 3,JK^a=0.538 3,JK^b=0.456 7,Sc1=1.000 0,Sc2=0,Co^a=0.992 4,Co^b=0.007 7,Lu^a=0,Lu^b=1.000 0,Au^a=0.849 5,Au^b=0.150 5。用χ~2检验比较基因型频率分布的观察值与期望值,符合Hardy-Weinberg遗传法则(P>0.05),新疆哈族人群稀有血型基因频率保持相对稳定;(2)发现哈萨克族人群有2个稀有血型系统基因表现型罕见存在,即S^-s^-2例,频率为0.010 2;Jk^(a-b-)1例,频率为0.005 1;(3)发现哈萨克族人群MNS、Kidd、Duffy、Dombrock和Diego 5种稀有血型系统基因频率分布呈现多态性,与其他民族人群明显不同,具有自身独特的遗传特点,而Kell、Colton、Scianna和Lutheran基因频率呈单态性分布特征。结论中国新疆哈萨克族人群9种稀有血型基因频率分布、遗传状况保持相对稳定,并存在独特的稀有基因型别。
Objective To study the distribution of blood group gene frequencies of MNS,Duffy,Kell,Dombrock,Diego,Kidd,Scianna,Colton and Lutheran in Kazakh populations of Xinjiang in order to provide evidence for the construction of popula鄄tion genetics,clinical blood and rare blood banks.Methods Nine blood groups of 196 Kazak ethnic groups in Xinjiang were geno鄄typed and analyzed by PCR-SSP.Results(1)The frequencies of nine rare blood group alleles in Kazak populations of Xinjiang were obtained:M=0.645 4,N=0.354 6,S=0.142 8,s=0.846 9,Fya=0.757 6,Fyb=0.242 3,K1=0.002 6,K2=0.997 5,Doa=0.221 9,Dob=0.778 0,Dia=0.035 7,Dib=0.964 3,JKa=0.538 3,JKb=0.456 7,Sc1=1.000 0,Sc2=0,Coa=0.992 4,Cob=0.007 7,Lua=0,Lub=1.000 0,Aua=0.849 5,and Aub=0.150 5.The frequency and distribution of genotypes were compared with Hardy-Weinberg rule(字2 test)(P>0.05).The gene frequency of rare blood groups in Kazakh populations remained stable.(2)There were rare phenotypes of rare blood groups in Kazak populations.The frequency of S-s-2 was 0.010 2,and that of Jka-b-1 was 0.005 1.(3)The distribution of gene frequencies of five rare blood groups of MNS,Kidd,Duffy,Dombrock and Diego was polymorphic,which was distinctly different from other ethnic groups.The gene frequency distribution of Kell,Colton,Scianna and Lutheran was monomorphic.Conclusion The genetic distribution and genetic status of nine rare blood groups in Kazak populations of Xinjiang are relatively stable with unique rare genotypes.
