摘要
从田间患软腐病的不结球白菜‘福冠’植株中分离纯化得到10个菌落,对引起软腐病的关键基因——果胶盐酸裂解酶(pel)基因的保守序列进行PCR扩增,测序发现该序列与胡萝卜果胶杆菌胡萝卜亚种(Pectobacterium carotovorum subsp.carovorum)果胶裂解酶基因(Gen Bank:J03673.1)的同源性达97.7%。对该菌株进行形态及生理生化分析,发现其符合软腐病菌特征特性。接种试验发现该菌株发病症状与田间不结球白菜软腐病一致。从接种病原菌的患病组织中重新分离病原菌并进行检测,发现其与所接种的病原菌为同种病原菌。因此,所分离出的病原菌为不结球白菜软腐病病原菌,且为胡萝卜果胶杆菌胡萝卜亚种。
Ten colonies were isolated and purified from soft rot diseased non-heading Chinese cabbage‘Fuguan’plants.The pectin hydrochloric acid lyase(pel)gene was amplified through PCR and followed by nucleotide sequencing.The pel sequence shared a 97.7%homology with the sequence of pectin lyase gene(GenBank:J03673.1)of Pectobacterium carotovorum subsp.carovorum.Morphological,physiological,and biochemical analyses of the strain was found to be consistent with the characteristics of soft rot bacterial pathogen.When the strain was inoculated into healthy non-heading Chinese cabbage,disease symptoms were produced that were consistent with the soft rot disease of non-heading cabbage in the field.The pathogens were re-isolated from the pathogen infected tissue and tested.It was found that the isolated pathogenic bacteria were the same as the inoculated pathogens.These results demonstrated that the pathogen of the non-heading cabbage soft rot disease was isolated and that it was the same as P.carotovorum subsp.carovorum.
