摘要
目的:分析丹皮酚对肝星状细胞活化及胶原合成的影响。方法:将人肝星状细胞分为空白组、模型组、丹皮酚低剂量组、丹皮酚高剂量组,加入不同试剂后,采用RT-PCR法、蛋白印迹法检测各组细胞COL1a1基因的表达,采用ELSIA法检测各组细胞培养上清液中羟脯氨酸的含量。结果:模型组COL1a1基因表达水平、羟脯氨酸含量明显高于其他三组(P<0.05),加入丹皮酚后人肝星状细胞COL1a1基因表达水平、羟脯氨酸含量开始下降(P<0.05)。并且其表达水平和含量在丹皮酚高剂量组低于丹皮酚低剂量组。结论:丹皮酚能够减弱TGF-β诱导的肝星状细胞的活化作用,并能抑制肝星状细胞胶原合成。
Objective:To analyze the effects of danpiol on the activation of hepatic stellate cells and collagen synthesis.Methods:human hepatic stellate cells can be divided into blank group,model group,low dose group were,high dose group,were to join different reagent,adopting rt-pcr method,protein imprinting method to detect each cell COLlal gene expression,The content of Hydroxypro-line in the cell culture supernatant of each group was detected by FLSLA.Results:the model group COLlal gene expression level and hydroxyproline content was significantly higher than the other three groups(P<0.05),to join the posterity hepatic stellate cells were COLlal gene expression level and hydroxyproline content began to decline(P<0.05).And the expression level and the content in the high dose group of danpiol were lower than danpiol low dose group.Conclusion:danpiphenol can reduce the activation of hepatic stel-late cells induced by TGF-beta,and can inhibit the synthesis of hepatic stellate cell collagen.
