淫羊藿多糖对气管切开插管留置大鼠肺部免疫功能的影响 被引量:1

The effects of Epimedium polysaccharides on pulmonary immune function in rats with tracheostomy

摘要 目的:探讨淫羊藿多糖对气管切开插管留置大鼠肺部免疫功能的影响。方法:将54只SD大鼠随机分为正常组(A组)、模型组(B组)、淫羊藿多糖组(C组),每组18只。B组和C组均复制气管切开插管大鼠模型,A组不做处理,6h后,C组灌胃给予淫羊藿多糖水溶液,A组、B组给予等量生理盐水。分别于灌胃后24h、72h、168h,每组各取6只大鼠,采用酶联免疫吸附试验(ELISA)法检测各组气管肺灌洗液中分泌型IgA(sIgA)和肺表面活性物质相关蛋白A(SP-A)含量及血清白介素-2(IL-2)水平。实时荧光定量PCR(qPCR)检测肺组织中rBD-2mRNA相对表达量。结果:(1)与A组比较,B组大鼠rBD-2mRNA相对表达量在造模后24h明显升高,随后逐渐下降,168h降到最低点(P<0.05);与B组比较,C组大鼠rBD-2mRNA相对表达量在造模后24h降低,72h、168h明显升高(P<0.05)。(2)与A组比较,B组大鼠肺灌洗液sIgA含量在造模后24h、72h明显升高(P<0.05),随后逐渐回落至正常水平(P>0.05);与B组比较,C组大鼠sIgA含量在造模后24h、168h明显升高(P<0.05)。(3)B组大鼠各时点SP-A含量均高于A组,但随着插管留置时间的延长,升高幅度逐渐降低(P<0.05);C组大鼠SP-A含量在造模后24h降低,但随着时间延长其含量逐渐升高,造模后168h明显高于B组(P<0.05)。(4)B组大鼠IL-2含量在造模后24h明显高于A组(P<0.05),与B组比较,C组IL-2含量在72h、168h明显升高(P<0.05),而两组在造模后24h无明显差异(P>0.05)。结论:淫羊藿多糖通过调节气管切开插管留置大鼠肺组织rBD-2mRNA表达,提高肺灌洗液sIgA、SP-A含量和外周血IL-2水平,从而改善气管切开插管留置大鼠肺部免疫功能。 54 SD rats were randomly divided into three groups(n=18 per group):a normal control group(group A),a model group(group B),and EPS group(group C).The rats in group B and group C were subjected to tracheotomy.After 6 h,rats in group C were treated with EPS via gavage,and those in groups A and B were treated with equal volume of saline.Secretory immunoglobulin A(sIgA)and pulmonary surfactant-related protein A(SP-A)levels in bronchoalveolar lavage fluid(BALF)and serum interleukin-2(IL-2)level were detected by enzyme-linked immunosorbent assay(ELISA).The mRNA expression ofβ-defensin-2(rBD2)mRNA in lung tissue was determined by real-time fluorescent quantitative PCR(qPCR).Results:(1)rBD-2 mRNA expression in group B was significantly higher than that in group A at 24 h after tracheostomy(P<0.05).Compared with group B,the expression of rBD-2 mRNA in group C was decreased at 24 h,while increased significantly at 72 h and 168 h(P<0.05).(2)The sIgA content in group B was significantly higher than that in group A after 24 h and 72 h of tracheotomy(P<0.05),and then gradually decreased to a normal level(P>0.05).Compared with group B,the content of sIgA in group C was notably elevated at 24 h and 168 h post-tracheotomy(P<0.05).(3)The content of SP-A in group B was higher than that in group A at each time point,but was gradually decreased as intubation time prolonged(P<0.05).The content of SP-A in group C was reduced at 24 h of modeling but increased with time prolonged,and was lower than that in group B at 168 h(P<0.05).(4)The level of IL-2 in group B was higher than that in group A at 24 h post-tracheotomy(P<0.05).Compared with group B,the serum IL-2 level in group C was markedly increased at 72 h and 168 h(P<0.05),but there was no significant difference in IL-2 level between groups B and C at 24 h(P>0.05).Conclusion:EPS could improve the pulmonary immune function of tracheostomy rats by regulating the rBD2 expression,elevate the contents of sIgA and SP-A in BALF and IL-2 level in serum.
