摘要
为了解迟缓爱德华菌鞭毛蛋白FliC的免疫特性,从迟缓爱德华菌基因组中克隆出鞭毛基因fliC,并将其构建到原核表达载体上,用原核表达的方法获得大量重组蛋白,将蛋白纯化后,通过动物模型确定该蛋白的免疫特性。结果表明,迟缓爱德华菌鞭毛蛋白FliC具有较强的免疫原性,对免疫动物应对强毒株感染具有一定保护效果。将FliC与牛血清白蛋白(BSA)混合免疫小鼠能够刺激机体产生较高水平抗BSA抗体,说明其佐剂特性。研究表明,迟缓爱德华菌鞭毛蛋白FliC具有优良的免疫原性和佐剂特性,是迟缓爱德华菌的保护性抗原,具有潜在的应用价值。
To investigate the immunological characters of flagellar protein FliC,in this study,fliC gene of Edwardsiella tarda was amplificated by PCR and cloned into pET-32a vector for for prokaryotic expression and purification,and abundant purified His-FliC was obtained.The recombinant FliC were then performed as immunogen for further immunological characters analysis.The data determined that,the FliC protein is able to sitmulate strong immune response and a certain specific protection to E.tarda infection.And the mice,received immunization combined FliC with BSA,displayed considerable anti-BSA serum antibody level,demonstrating the adjuvant characteristics of FliC.All these work showed that the flagellar protein FliC of E.tarda possess excellent immunogenicity and adjuvant characteristics,and may be a good protective antigen of E.tarda for vaccine development.
